Degumming process and silk properties of tussah cocoon using Esperase
Esperase protease with high activity and a broad substrate spectrum is used as a biology degumming agent,the effects of enzyme dosage and enzyme treatment time on the degumming rate of tussah cocoon are investigated,and the enzymatic degumming process is optimized.The physicochemical properties of degumming solution and degumming silk fibroin are characterized using SDS-PAGE,scanning electron microscopy(SEM)and fiber strength meter,and compared with those of sodium carbonate degumming method.The results show that the optimum degumming conditions are 100 μL enzyme dosage per gram of tussah cocoon,treating at 50 ℃ for 2 h.Under the optimal degumming conditions,the degumming rate is 12.83%,sericin is hydrolyzed into small molecules with a molecular weight less than 10 000 Da;the surface of silk fibroin is smooth and flat,the breaking strength of degummed silk slightly decreases while the toughness improves.Compared with alkali degumming,enzymatic degumming method with esperase has a more uniform degumming effect.
