越来越多的研究表明,动脉粥样硬化(atherosclerosis,AS)的发生与细胞焦亡特别是GSDMD (gasdermin D)介导的细胞焦亡有着密切的关系.AS中巨噬细胞、血管内皮细胞和血管平滑肌细胞等炎性细胞发生细胞焦亡导致斑块破裂引起血栓形成,从而引发急性冠状动脉综合征.在AS发展的过程中,动脉管壁内皮损伤和脂质沉积是主要的始动因素,它分别通过Caspase-1和Caspase-11/4/5的激活对GSDMD蛋白进行切割,介导经典和非经典细胞焦亡.GSDMD蛋白的N端迁移至细胞膜上,然后寡聚化在细胞膜上形成孔道,释放炎性因子IL-1β和IL-18到细胞间质中.炎性因子的释放进一步促进了血管内皮细胞的焦亡,细胞焦亡后其内容物、细胞因子和蛋白酶进入到细胞外基质中,加重了局部炎症反应,导致AS斑块破裂和心血管事件的发生.基于上述机制,直接靶向GSDMD以及靶向GSDMD诱导的细胞焦亡信号通路中的信号分子如炎性小体、Caspase-1、 IL-1β和IL-18等的药物开发,为AS的治疗提供了理论依据和新思路.
Research progress on the role of GSDMD-mediated pyroptosis in athero-sclerosis
More and more studies have shown that the occurrence of atherosclerosis is closely related to pyroptosis,especially gasdermin D(GSDMD)-mediated pyroptosis.In atherosclerosis,inflammatory cells such as macrophages,vascular endothelial cells,and vascular smooth muscle cells undergo pyroptosis,leading to plaque rupture and thrombosis,and finally to acute cor-onary syndrome.In the development of atherosclerosis,arterial wall endothelial injury and lipid deposition are the main initia-ting factors,which activate Caspase-1 and Caspase-11/4/5,respectively,and in turn cause the cleavage of the GSDMD protein and mediate classical and non-classical cell pyroptosis.The N-terminus of the GSDMD protein migrates to the cell membrane and oligomerizes to form pores through which the inflammatory factors IL-1β and IL-18 are released into the interstitium.The release of inflammatory factors further promotes the death of intravascular cells,after which their contents,cytokines,and proteases enter the extracellular matrix,aggravating the local inflammatory response,leading to atherosclerotic plaque rupture and cardiovascular disease.Based on the above mechanism,the development of drugs that directly target GSDMD and signal molecules in the pyroptosis signaling pathway induced by GSDMD,such as inflammasomes,Caspase-1,IL-1β,and IL-18,provides a theoretical basis and new ideas for the treatment of atherosclerosis.
