TLR属于模式识别受体家族中的一员,能够识别多种类型的PAMP.TLR信号通路在B细胞分化和激活过程中起着多种作用.为研究zeste基因的增强子同源物2(enhancer of zeste homolog 2,EZH2)在B细胞TLR介导的免疫反应中的作用,调取国家基因组科学数据中心与人类B细胞TLR信号通路有关的转录组数据,初步分析多梳抑制复合体2(polycomb repressive complex 2,PRC2)亚基的表达改变.随后分离人PBMC中的B细胞,用qRT-PCR和Western blotting的方法从转录水平和蛋白水平检测B细胞TLR通路激活后EZH2的表达水平.结果显示B细胞TLR激活后EZH2的表达增加.接着用EZH2抑制剂3-去氮腺嘌呤(3-deazaneplanocin A,DZNep)预处理人PBMC细胞,用流式细胞术和ELISA检测TLR介导的B细胞增殖和抗体分泌的改变.结果显示抑制B细胞中EZH2的功能后,TLR9介导的B细胞增殖和抗体分泌减少.综上,EZH2能够参与B细胞TLR介导的免疫反应,抑制B细胞中EZH2的表达后,TLR介导的B细胞增殖和抗体分泌减少.
EZH2 is involved in TLR-mediated B cell proliferation and antibody secretion
TLR is a member of the pattern recognition receptor family,which recognizes many types of PAMP.TLR signaling plays multiple roles in B-cell differentiation and activation.To investigate the role of enhancer of zeste homolog 2(EZH2)in TLR-mediated immune response of B cells,transcriptomic data related to TLR signaling pathway of human B cells were col-lected from National Genome Science Data Center and expression changes of polycomb repressive complex 2(PRC2)subunits were analyzed.EZH2 expression in PBMC after activation via TLR pathway was detected by qRT-PCR and Western blotting.The results showed that EZH2 expression increased after TLR activation in B cells.Next,human PBMC were pretreated with EZH2 inhibitor 3-deazaneplanocin A(DZNep),and TLR-mediated changes in B cell proliferation and antibody secretion were detected by flow cytometry and ELISA.The results showed that inhibition of EZH2 in B cells resulted in a decrease in TLR9-mediated B cell proliferation and antibody secretion.In conclusion,EZH2 participates in the TLR-mediated immune response of B cells.Inhibition of EZH2 expression in B cells leads to a decrease in TLR-mediated B cell proliferation and antibody secretion.
