Establishment of Technological System for In Vitro Propagation of Dendrobium aduncum
In order to accelerate the breeding process, rescue rare and endangered variety and provide the premise for in vitro propagation of rare plant Dendrobium, in this study, the immature seeds used as explants were applied to conduct rapid in vitro breeding of Dendrobium aduncum. The results showed that the protocorm was induced directly on MS medium containing 0. 5~1. 0 mg/L of BA and 0. 2 mg/L of NAA. The optimal medium for proctocorm multiplication was 2. 0 mg/L MS + BA + 0. 5 mg/L KT+0. 5 mg/L NAA with the propagation coefficient up to 12. 4. The MS medium supplemented with 0. 5 mg/L BA,0. 5 mg/L KT and 0. 2 mg/L NAA was suitable for the differentiation of the adventitious buds with the differentiation percentage of 97. 5%. The optimal medium for root formation was MS 1/2 + 0. 5 mg/L IBA+0. 5 mg/L NAA. The number of root per plantlet was 8. 57 and the rooting rate up to 100%. The plantlets grew well after transplanted.
