猪源化脓隐秘杆菌的分离鉴定及病理组织学观察

摘要
从吉林省某猪场1例病死猪肺脏内分离到1株革兰氏阳性杆菌,该菌接种于TSA(含5％胎牛血清)平板,培养18 h长出肉眼可见的菌落,而在巧克力平板上生长缓慢.对该菌株的16S rRNA基因进行PCR扩增并进行序列测定,测序结果经Nast分析与化脓隐秘杆菌的核苷酸序列同源性为99％,经过进一步糖、醇发酵试验及触酶试验等一系列生化试验,结果与测序结果相符,将其命名为AP4.该菌对供试小鼠具有致病性.药敏试验结果显示,该菌对氯霉素、氟苯尼考、阿米卡星及庆大霉素敏感,对红霉素、阿奇霉素、环丙沙星、强力霉素、阿莫西林及磺胺间甲氧嘧啶耐药.
Abstract
A small gram-positive strain was isolated from the lung of dead swine.Colonies could be seen growing well on TSA plate(containing 5％fetal calf serum)but poorly on chocolate agar plate after 18 hours of culture.Its 16S rRNA gene was cloned with PCR and then sequenced.Blast result showed that the sequence was 99％ homologous with the 16S rRNA gene of Arcanobacterium pyogenes.The sugar,alcohol fermentation experiments,catalase test and other biochemical test results were consistent with the sequencing result,therefore the strain was named AP4.The isolated strain had pathogenicity to mice.Drug sensitivity test results showed that the strain was sensitive to antibiotics of florfenicol,chloramphenicol,amikacin and gentamicin,but resistant to erythromycin,azithromycin,ciprofloxacin,amoxicillin and sulfamonomethoxine.
