低温胁迫对番茄RNA解旋酶SlDEAD34基因表达的影响

利用生物信息学方法和实时荧光定量PCR技术对番茄RNA解旋酶SlDEAD34的蛋白结构特征、SlDEAD34基因的组织表达模式和低温胁迫条件下的基因表达情况进行了研究.结果表明,SlDEAD34包含9个保守基序,是DEAD-box型RNA解旋酶,与拟南芥LOS4高度同源,三维结构非常类似;SlDEAD34基因在生长6周的番茄根、茎和叶中均有表达,且根>茎>叶,在生长5个月的番茄9种组织器官中均有表达,其中在果实中表达量最高,在根中表达量最低;4℃处理时,番茄根中SlDEAD34基因下调明显,叶中表达量变化不明显,12℃处理时,番茄根和叶中SlDEAD34基因表达量变化均不明显.DEAD-box型RNA解旋酶SlDEAD34经低温4℃处理后基因表达量明显下调,暗示SlDEAD34基因响应低温胁迫,推测SlDEAD34基因可能参与调控低温逆境胁迫过程.
Gene Expression Analysis of RNA Helicase Gene SIDEAD34 under Low Temperature Stress in Tomato
Bioinformatics and quantitative RT-PCR were employed to study the protein structure of SlDEAD34 and the expression pattern of SlDEAD34 in different vegetative organs under low temperature stress in tomato.Results showed that SlDEAD34,highly homologous with Arabidopsis LOS4,is a DEADbox RNA helicase with 9 conserved motifs.SlDEAD34 expresses in all tested tissues of 6 weeks seedling,with the highest level in root and lowest in leaf,whereas with the highest level in fruit and lowest in root of 5 months mature plants.Interestingly,the expression levels of SlDEAD34 in root were much lower than in leaf under the low temperature stress of 4 ℃.However,the gene expression of SlDEAD34 has no obvious change in both root and leaf under 12 ℃ treatment in tomato.In conclusion,SlDEAD34 was downregulated significantly after low temperature 4 ℃ treatment.Taken together,we speculated that SlDEAD34 might be involved in the response of tomato to low temperature stress.
