单核细胞増生李斯特菌LIPI-4对脑及脑微血管内皮细胞炎症小体表达的影响

目的 探究单核细胞増生李斯特菌毒力岛 4(LIPI-4)对小鼠脑及脑微血管内皮细胞炎症小体产生的影响.方法 小鼠腹腔注射细菌 Lm928 和 Lm928ΔLIPI-4,采用甲酞胺一紫外分光光度法检测脑组织中伊文思蓝(EB)的含量,ELISA方法检测小鼠血清中髓鞘碱性蛋白(MBP)的水平,qRT-PCR 法检测脑组织中炎症小体 AIM2、NLRP3、NLRC4,炎症因子 IL-1β、IL-18 等的表达.qRT-PCR、Western Blot 和 ELISA 检测 Lm928、Lm928ΔLIPI-4 和CLm928ΔLIPI-4 侵染人脑微血管内皮细胞(HCMEC/D3)过程中炎症小体相关分子及炎症因子表达的水平.结果 小鼠感染 Lm92836 h后脑内 EB 显著升高,72 h 后,血清中 MBP 含量极显著高于 Lm928ΔLIPI-4 组(P<0.01),qRT-PCR结果显示 Lm928 组小鼠脑组织 NLRC4 和 IL-1β的 mRNA表达量显著高于 Lm928ΔLIPI-4 组(P<0.05).在侵染 HCMEC/D3 细胞中,NLRP3 和 IL-1β的 mRNA 表达量,Lm928 组极显著高于 Lm928ΔLIPI-4 组(P<0.01);NLRP3 的蛋白表达量,Lm928 组极显著高于Lm928ΔLIPI-4 组(P<0.01);上清液中IL-1β的分泌量,Lm928 组极显著高于 Lm928ΔLIPI-4 组(P<0.01).结论 单核细胞增生李斯特菌 LIPI-4 通过参与 NLRP3/NLRC4/IL-1β 通路,激活炎症小体参与炎症反应,损伤脑组织与脑微血管内皮细胞,破坏血脑屏障.
Effects of Listeria monocytogenes LIPI-4 on the expression of inflammasome in brain and brain microvascular endothelial cells
ObjectiveTo explore the effect of Listeria monocytogenes Pathogenicity island 4(LIPI-4)on the expression of inflamma-some in brain microvascular endothelial cells.Methods Mice were intraperitoneally injected with bacteria Lm928 and Lm928ΔLIPI-4.The content of Evans blue(EB)in brain tissue was detected by methylphthalamide ultraviolet spectrophotometry,the level of Myelin basic protein(MBP)in serum was detected by ELISA,and the expression of NLRP3,AIM2,NLRC4,inflammatory factors IL-1β,IL-18,etc.in brain tissue was detected by qRT-PCR.qRT-PCR,Western Blot,and ELISA were used to detect the inflammasome re-lated molecules and inflammatory factors expression of human brain microvascular endothelial cells(HCMEC/D3)infected by Lm928、Lm928ΔLIPI-4 and CLm928ΔLIPI-4.Results After 36 h of infection with Lm928,EB content in the mouse brain significantly in-creased.After 72 h of infection with Lm928,the serum MBP content was significantly higher than that of the Lm928ΔLIPI-4 group(P<0.01).The qRT-PCR results showed that the mRNA expression levels of NLRC4 and IL-1β in the Lm928 group were significant-ly higher than those in the Lm928ΔLIPI-4 group(P<0.05).In infected HCMEC/D3 cells,the mRNA expression levels of NLRP3 and IL-1β in the Lm928 group were significantly higher than those in the Lm928ΔΔLIPI-4 group(P<0.01);The Western blot re-sults showed that the protein expression levels of NLRP3 and NLRC4 in the Lm928 group were significantly higher than those in the Lm928ΔLIPI-4 group(P<0.01);The secretion of IL-1β in the HCMEC/D3 supernatant of the Lm928 group was significantly higher than that of the Lm928ΔLIPI-4 group(P<0.01).Conclusion Listeria monocytogenes LIPI-4 participates in the NLRP3/NLRC4/IL-1β pathway,activates inflammasomes to participate in inflammatory responses,damages brain tissue and brain microvascular endotheli-al cells,and destroys the blood-brain barrier.
