摘要
目的 研究 Ⅱ 型钙粘蛋白 18(Cadherin 18,CDH18)在宫颈癌中的表达及其对宫颈癌 SiHa、HeLa和 C33A细胞增殖和迁移能力的影响.方法 采用qRT-PCR、Western blot实验检测宫颈癌组织和细胞中 CDH18 的mRNA和蛋白质表达水平;分别构建过表达和敲低 CDH18 基因表达的质粒;在稳定转染质粒的宫颈癌细胞中,利用 CCK-8、Tran-swell和划痕实验分析过表达及降低 CDH18 基因对SiHa、HeLa和C33A细胞增殖和迁移的影响.结果 高危型HPV阳性的宫颈组织和细胞中 CDH18 蛋白质和 mRNA 表达水平均显著低于正常宫颈组织和细胞(P<0.01),差异具有统计学意义;CCK-8、Transwell 及划痕实验结果显示:过表达 CDH18 基因促进SiHa、HeLa和C33A细胞的增殖和迁移能力(P<0.01);敲低 CDH18 基因抑制了SiHa、HeLa和C33A细胞的增殖和迁移能力(P<0.05).结论 CDH18 基因在高危型 HPV阳性宫颈癌组织和细胞中低表达,在高危型 HPV 阴性宫颈癌组织细胞中高表达;其过表达能够促进宫颈癌细胞的增殖和迁移.
Abstract
Objective To investigate the expression of Cadherin 18(CDH18)in cervical cancer and its effect on the proliferation and migration of SiHa,HeLa and C33A cells.Methods The mRNA and protein expression levels of CDH18 in cervical cancer tissues and cells were individually detected by qRT-PCR and Western blot,respectively.Plasmid with overexpression and the counterpart with knockdown CDH18 gene expression were constructed respectively.The effects of overexpression and knockdown of CDH18 gene on the proliferation and migration of SiHa,HeLa and C33A cells were analyzed by CCK-8,Transwell and scratch assay.Results The expres-sion levels of CDH18 protein and mRNA in high-risk HPV positive cervical tissues and cells were significantly lower than those in nor-mal cervical tissues and cells(P<0.01),and the differences were statistically significant.The results of CCK-8,Transwell and scratch tests showed that overexpression of CDH18 gene promoted the proliferation and migration ability of SiHa,HeLa and C33A cells(P<0.01).Knockdown of CDH18 inhibited the proliferation and migration of SiHa,HeLa and C33A cells(P<0.05).Conclusion The expression of CDH18 gene is low in high-risk HPV positive cervical cancer tissues and cells,it is highly expressed in high-risk HPV-negative cervical cancer cells;Its overexpression can promote the proliferation and migration of HPV positive cervical cancer cells.
