Effect of CDH18 gene on proliferation and migration of cervical cancer cells
Objective To investigate the expression of Cadherin 18(CDH18)in cervical cancer and its effect on the proliferation and migration of SiHa,HeLa and C33A cells.Methods The mRNA and protein expression levels of CDH18 in cervical cancer tissues and cells were individually detected by qRT-PCR and Western blot,respectively.Plasmid with overexpression and the counterpart with knockdown CDH18 gene expression were constructed respectively.The effects of overexpression and knockdown of CDH18 gene on the proliferation and migration of SiHa,HeLa and C33A cells were analyzed by CCK-8,Transwell and scratch assay.Results The expres-sion levels of CDH18 protein and mRNA in high-risk HPV positive cervical tissues and cells were significantly lower than those in nor-mal cervical tissues and cells(P<0.01),and the differences were statistically significant.The results of CCK-8,Transwell and scratch tests showed that overexpression of CDH18 gene promoted the proliferation and migration ability of SiHa,HeLa and C33A cells(P<0.01).Knockdown of CDH18 inhibited the proliferation and migration of SiHa,HeLa and C33A cells(P<0.05).Conclusion The expression of CDH18 gene is low in high-risk HPV positive cervical cancer tissues and cells,it is highly expressed in high-risk HPV-negative cervical cancer cells;Its overexpression can promote the proliferation and migration of HPV positive cervical cancer cells.
