The effect of Yuxuebi tablet on rheumatoid arthritis based on intestinal flora
Objective To explore the effect of Yuxuebi tablet in the treatment of rheumatoid arthritis based on intestinal flora.Methods Thirty-six SD rats were randomly divided into normal control group,collagen induced rheumatoid arthritis(CIA)group,methotrexate 0.002 g·kg-1 dose group,Yuxuebi tablet 0.4,0.8,1.6 g·kg-1(YXBL,YXBM,YXBH)dose groups,the latter five groups were treated with bovine type Ⅱ collagen emulsion to induce rheumatoid arthritis model.Except for rats in the normal group and CIA group,which were given three distilled water,the other groups were given the above doses by intragastric administration.After 30 days,the weight of spleen and thymus of rats in each group was measured,and the spleen index and thymus index were calculated.HE staining was used to observe the pathological changes of synovial tissue,spleen and colon.16S rRNA high-throughput sequencing technology was used to detect the changes in intestinal flora of rats in each group.Results The spleen index and thymus index of CIA rats were re-duced(P>0.05),the paw volume,ankle circumference and arthritis score were decreased(P<0.01),the synovial tissue hyperplasia and inflammatory cell infiltration were reduced,and the pathological changes of spleen and colon were improved after the CIA rats were treated with Yuxuebi tablet.Yuxuebi tablet could reduce the relative abundance of Fusicatenibacter,Erysipelotrichaceae_UCG-003,Monoglobus and UCG-005(P<0.05),and increase the relative abundance of Prevotellaceae_UCG-003 in the intestinal flora of CIA rats(P<0.05).Conclusion Yuxuebi tablet can improve rheumatoid arthritis,which may be related to its regulation of intestinal flora.
