Construction of LncRNA-related CeRNA network for intramuscular fat formation in chicken embryo
Objective The aim of this study was to screen the differential expression of long non-coding RNA(lncRNA),microRNA(miRNA)and mRNA in chicken embryonic muscle.And to explore the important candidate genes of intramuscular fat formation by constructing lncRNA-miRNA-mRNA competitive regulatory network.Methods In this study,the muscle tissues of Rose Crown chicken and Cobb chicken at embryonic period were collected for transcriptome sequencing,and the differentially expressed lncRNA,miRNA and mRNA were screened by bioinformatics methods.We analyzed the GO enrichment and KEGG signal transduction pathway of differ-entially expressed genes,and then predicted the target relationship between lncRNA-miRNA and miRNA-mRNA.Combined with gene expression analysis,a lncRNA-miRNA-mRNA competitive regulatory network that may be related to the formation of intramuscular fat was constructed,and used qRT-PCR to verify the accuracy of the sequencing data.Results The results showed that there were 5 lncR-NAs,225 miRNAs and 146 mRNAs common differentially expressed between breast and leg muscles of Rose crowned chicken and Cobb chicken.GO and KEGG results showed that the target genes were enriched in the signaling pathways related to lipid metabolism or adipogenesis.A 16-node ceRNA regulatory network consisting of 4 lncRNAs,6 miRNAs and 6 mRNAs was constructed.Conclu-sions The ceRNA regulatory network of chicken intramuscular fat formation was successfully constructed.This study provided a refer-ence for further exploring the molecular regulation mechanism of intramuscular fat formation.And laid a foundation for the breeding of high-quality broilers in the future.
