摘要
为了筛选可应用于香猪用饲料添加剂的乳酸菌菌株,试验通过1%碳酸钙的MRS固体培养基从香猪盲肠内容物中分离筛选出1株产酸能力强、抗逆性好、抗生素敏感的乳酸菌.经革兰氏染色鉴定、生理生化鉴定、16S rRNA测序,鉴定该乳酸菌为植物乳杆菌(L.plantarum).进一步从产酸性能、耐酸试验、耐热试验、耐胆盐试验、抗生素敏感性试验等方面对分离到的L.plantarum进行生物学特性研究.结果表明:L.plantarum生长最适温度为37~45℃;L.plantarum具有优秀的产酸能力,pH最低可达到3.3;L.plantarum可以耐受低pH,在pH为2.0时的存活率为13%;在含0.4%猪胆盐的MRS肉汤中存活率为13%;L.plantarum对红霉素、复方新诺明、氯霉素、青霉素、氨苄西林、头孢唑林高度敏感.由此可见,L.plantarum是一株抗逆性、抗生素敏感性、产酸效果都非常优异的香猪源微生态制剂备用菌株.
Abstract
In order to screen lactic acid bacteria strains that can be applied as feed additives for fragrant pigs,this experiment isolated and screened a lactic acid bacteria strain with strong acid production abil-ity,good stress resistance,and antibiotic sensitivity from the cecum content of fragrant pigs using 1%calcium carbonate MRS solid culture medium.After Gram staining,physiological and biochemical identi-fication,and 16S rRNA sequencing,the lactic acid bacteria were identified as L.plantarum.Further analysis was conducted on the isolated L.from aspects such as acid production L.performance,acid resis-tance test,heat resistance test,bile salt resistance test,and antibiotic sensitivity test Plantarum conducts research on biological characteristics.The results indicate that L The optimal temperature for plantarum growth is 37-45℃.L.Plantarum has excellent acid production ability,with a minimum pH of 3.3.L.Plantarum can tolerate low pH and has a survival rate of 13%at a pH of 2.0.The survival rate in MRS broth containing 0.4%pig bile salt was 13%.L.Plantarum is highly sensitive to erythromycin,compound sulfamethoxazole,chloramphenicol,penicillin,ampicillin,and cefazolin.From this,it can be seen that L.Plantarum is a backup strain of probiotics derived from fragrant pigs with excellent stress resistance,an-tibiotic sensitivity,and acid production efficiency.
