Optimization of Total Organic Acid Extraction Process and Antioxidant Activity of Fructus Cinnamomi Cassiae Immaturi by Response Surface Methodology
To optimize the extraction process of total organic acids from Fructus Cinnamomi cassiae im-maturi using response surface methodology and study their in vitro antioxidant activity.Based on the de-sign principle of Box Behnken Design,the effects of ethanol concentration,solid-liquid ratio,ultrasound power,and ultrasound duration on the total organic acid yield of cinnamon seeds were investigated.Using the yield of total organic acids from Fructus Cinnamomi cassiae immaturi as the response value,the opti-mal extraction process for total organic acids from Fructus Cinnamomi cassiae immaturi was determined through response surface optimization.DPPH method and iron ion reduction ability were used to evaluate the total organic acid antioxidant capacity of Fructus Cinnamomi cassiae immaturi.The results indicate that the order of single factor influence is material liquid ratio>ethanol concentration>ultrasonic time.The opti-mal extraction process for total organic acids in Fructus Cinnamomi cassiae immaturi is as follows:ethanol concentration 59%,solid-liquid ratio 1∶60(g/mL),ultrasonic power 350 W,ultrasonic time 49 minutes,and the average content of total organic acids ob-tained is 9.77%.The IC50 of VC and S-1 were 0.011 19 mg/mL and 0.014 37 mg/mL,respec-tively.This extraction process is stable,feasible,and reliable;It has certain antioxidant capacity.
