Optimization of Extraction Process of Total Triterpenoids from Passiflora edulis Sims Shell and Evaluation of Antioxidant Activity
The purpose of this study was to provide reference for the research of active components of Passi-flora edulis Sims shell and the development of functional products.With ursolic acid as the standard,the con-tent of total triterpenoids(PESTT)in Passiflora edulis Sims shell was determined by vanillin glacial acetic acid perchloric acid colorimetry.On the basis of single factor experiment,response surface methodology was used to optimize the extraction conditions of PESTT,and the antioxidant capacity of PESTT was evaluated by determining the scavenging rates of 1,1-diphenyl-2-trinitrophenylhydrazine free radical(DPPH·)and hydroxyl free radical(·OH).The results showed that the optimal extraction conditions of PESTT were opti-mized by response surface methodology as follows:ethanol volume fraction 87%,ultrasonic temperature 43℃,ultrasonic time 46 min,liquid-solid ratio 33∶1(mL/g),ultrasonic power 300 W,under the optimal conditions,the average extraction amount of PESTT was(6.01±0.16)mg/g,which was very close to the model prediction value(5.97 mg/g);when the mass concentration of PESTT was 0.005 mg/mL,the clearance rates of DPPH·and·OH were 74.4%and 86.2%,respec-tively,and the clearance rate was higher than that of the control drug vitamin C.Response surface methodology was feasible to optimize the extrac-tion conditions of PESTT.PESTT had strong anti-oxidant activity,and Passiflora edulis Sims shell could be used to develop functional products.
