The aim of this study was to optimize the toxicity of zearalenone(ZEN)and to explore the effect of ZEN on the estrogen response of zebrafish.By changing the medium formula and using high performance liquid chromatography(HPLC),it was found that an environment that provides inorganic salts required for growth is conducive to the accumulation of ZEN.In the toxicity test,after zebrafish embryos were exposed to different concentrations of ZEN(0,0.1,0.2,0.4 μg/mL),the heart rate of zebrafish decreased with increasing concentration,the hatching rate of embryos at 48 h and 72 h was significantly inhibited,and deformities such as spinal curvature,tail curvature,and yolk sac edema occurred.At the same time,the expression of estrogen in embryos exposed to ZEN for 72 h was significantly inhibited after 72 h of exposure,and it showed a concentration gradient effect.Further experiments showed that exposure to 0.2 μg/L ZEN for 7 days significantly affected the fertilization rate and embryonic survival rate of adult zebrafish,and the effect on female fish was greater than that on male fish.The results clearly showed that the accumulation of ZEN was related to the addition of inorganic salts in the culture medium,indicating that ZEN was significantly toxic to the embryonic development of zebrafish and that its reproductive toxicity to adult zebrafish was significantly gender-biased,which could provide a scientific basis for the detoxification,detection and environmental management of ZEN.
