摘要

目的:探讨茯苓水提取物、三七水提取物及其提取物对小鼠睡眠作用的影响及作用机制.方法:采用直接睡眠、戊巴比妥钠延长睡眠时间、戊巴比妥钠阈下剂量催眠、巴比妥钠睡眠潜伏期,检测小鼠下丘脑中神经递质含量的变化,评价其对小鼠睡眠情况的影响,通过急性毒性实验评价茯苓三七水提取物复合物的安全性.结果:茯苓水提取物、三七水提取物及其水提取物复合物各剂量组结果显示,戊巴比妥钠诱导可以延长小鼠的睡眠时间.茯苓三七水提取物复合物各剂量组小鼠睡眠发生率显著提高,差异有统计学意义(P<0.05),除三七水提取物低剂量组外,其余各组小鼠的睡眠潜伏期有所缩短,差异有统计学意义(P<0.05).此外,茯苓三七水提取物复合物中、高剂量组小鼠下丘脑及血清中的5-羟色胺(5-HT)与γ-氨基丁酸(GABA)含量均显著高于对照组,差异有统计学意义(P<0.05),明显增加失眠大鼠下丘脑中的5-HT和GABA含量,调节下丘脑中神经递质含量的变化,并且不会产生毒性.结论:茯苓水提取物、三七水提取物及茯苓三七水提取物复合物均可通过调节神经递质等方面改善小鼠睡眠,并且茯苓水提取物和三七水提取物组成的复合物具有协同增效,安全性良好.

Abstract

Objective:To explore the effects and mechanisms of Poria cocos water extract,Panax notoginseng water extract and their extracts on sleep in mice.Methods:Direct sleep,prolonged sleep time with pentobarbital sodium,subthreshold dose hypnosis with pentobarbital sodium,and sleep latency with pentobarbital sodium were used to detect the changes in neurotransmitter content in the hypothalamus of mice,evaluate its impact on mouse sleep,and evaluate the safety of Poria cocos notoginseng water extract complex through acute toxicity experiments.Results:The results of each dose group of Poria cocos water extract,Panax notogin-seng water extract and their water extract complex showed that pentobarbital sodium induction can prolong the sleep time of mice.The sleep incidence of mice in each dose group of Poria cocos and Panax notoginseng water extract complex was significantly increased,and the difference was statistically significant(P<0.05).Except for the low-dose group of Panax notoginseng water ex-tract,the sleep latency of other groups of mice was shortened,and the difference was statistically significant(P<0.05).In addi-tion,the levels of 5-hydroxytryptamine(5-HT)in the hypothalamus and serum of mice in the middle and high-dose groups of Poria cocos and Panax notoginseng water extract complex were found to be correlated with-γ-Aminobutyric acid γ-The content of Ami-nobutyric acid(GABA)was significantly higher than that of the control group,and the difference was statistically significant(P<0.05).It significantly increased the content of 5-HT and GABA in the hypothalamus of insomnia rats,regulated the change of the content of neurotransmitters in the hypothalamus,and did not produce toxicity.Conclusion:Pachycoia water extract,Notoginseng water extract and pachycoia Notoginseng water extract complex can improve sleep of mice by regulating neurotransmitter,and the complex composed of pachycoia water extract and Notoginseng water extract has synergistic effect and good safety.
