摘要
目的:初步探讨匝迪-5味丸对抑郁模型大鼠行为学及学习记忆能力的影响,评价其抗抑郁作用及对学习记忆的干预作用。方法 SD 雄性大鼠,根据体质量随机分为正常组、模型组、氟西汀组(3.6 mg/ kg),匝迪-5味丸低、中、高剂量组(0.2、0.4、0.8 mg/ kg),每组10只。采用应激结合孤养的方法复制抑郁模型,以矿场(Open - field)实验、糖水消耗实验及体质量变化来进行行为学评价,应用 Morris 水迷宫试验来检测大鼠学习记忆能力。结果给予21d 应激后模型大鼠糖水消耗量减少,体质量增长缓慢,Open - Field 实验水平活动得分和垂直活动得分及 Morris 水迷宫实验逃避潜伏期较正常组显著降低(P <0.05,P <0.01)。与模型组相比,氟西汀和匝迪-5味丸高、中剂量组可不同程度提高模型大鼠糖水消耗量、Open - Field 实验水平活动和垂直活动及学习记忆能力( P <0.05,P <0.01)。结论匝迪-5味丸具有一定的抗抑郁作用,其机制值得进一步研究。
Abstract
Objective To preliminary study the effect of Zadi Wuwei pills for the ability of learning and memory behavior on depression model rats,and to evaluate its intervention effect of its antidepressant effects of learning and memory. Methods According to weight,male SD rats were randomly divided into nor-mal group,model group,positive control group(3. 6 mg/ kg),Zadi Wuwei pills low,medium and high dose groups(0. 2,0. 4,0. 8 mg/ kg),and there were 10 rats in each group. The stress method combined with isola-ted raising were used to replicate model of depression rats,and mine(Open - field)test,sucrose consumption test and weight change were used to evaluate behavior. Morris water maze test was applied to test the learning and memory ability of rats. Results After 21 days of stress method,sugar water consumption decreased,and there was slow increase of body weight. Open - Field level test activity scores and vertical activity score and Morris watermaze test escape latency were significantly lower than those of the normal group(P < 0. 01 or 0. 05). Compared with model group,fluoxetine and Zadi Wuwei pills,in high dose increased the sugar con-sumption,and Open - Field test of horizontal and vertical activity and the ability of learning and memory showed better results(P < 0. 01 or 0. 05). Conclusion Zadi Wuwei pills has certain antidepressant effect, and it is worth of further study on mechanism.
