Study on the Therapeutic Effect of Qizhiweitong Granule on Acute Gastritis in Rats
Objective To explore the therapeutic effect of Qizhiweitong Granule on acute gastritis in rats and pre-liminarily reveal its mechanism of action.Methods A total of 56 male SPF Wistar rats were randomly divided into blank group,model group,positive group and low-,medium-and high-dose groups of Qizhiweitong Granule,with 8 rats in each group.Acute gastritis in rats was induced by 70％ ethanol,and after drug treatment,the pathological changes in rat gastric tissue were observed using HE staining.The contents of TNF-α,SOD and TGF-β in serum were detected by ELISA,and the expression levels of INOS,HIF-1α and IFNG in gastric tissue were determiend by Western blot.Results After the treatment with Qizhiweitong Granule,the gastric bleeding spots in rats with acute gastritis were significantly im-proved compared with the model group,with gradual recovery of gastric folds and a brighter color.The structure of gastric tissue was basically intact,with a slight loss of chief cells,a few blood cells in the interstitial space,no inflammatory cell infiltration,and overall structural integrity.The contents of TNF-α and TGF-β in serum of rats were significantly de-creased(P＜0.05),while the content of SOD was significantly increased(P＜0.05).The expressions of INOS,HIF-1αand IFNG(IFN-γ)in gastric tissues were significantly decreased(P＜0.05).Conclusion Qizhiweitong Granule have an excellent therapeutic effect on acute gastritis in rats,and its mechanism of action is related to the HIF-1 signaling path-way.
