目的 基于生物信息学技术,筛选溃疡性结肠炎(Ulcerative colitis,UC)关键免疫靶基因,分析相关机制,筛选有效治疗中药,为UC基础研究及中医药精准辨证治疗提供理论支持.方法 应用GEO数据集、ImmPort数据库、SRTING数据库、cytoHubba插件等筛选UC关键免疫靶基因.对靶基因进行logistic单因素分析,应用cor函数进行相关性分析,应用CIBERSORT算法进行免疫浸润分析,应用Enrichr数据库进行KEGG、GO富集分析,通过miRWalk、RNA22、RNAInter、TargetScan数据库检索筛选miRNA,并通过Coremine Medical数据库筛选潜在治疗中药.结果 筛选出15个靶基因呈表达上调,均为UC独立危险因素.免疫浸润分析显示,CD4+记忆性T细胞、M0巨噬细胞、M1巨噬细胞、中性粒细胞等在组间差异表达,且靶基因与免疫细胞相关性密切.KEGG分析IL-17信号通路、TNF信号通路等是重要通路,GO分析生物过程主要在中性粒细胞、粒细胞趋化作用等方面,细胞成分主要在三级颗粒管、含胶原蛋白的细胞外基质等方面,分子功能主要在趋化因子活性、趋化因子受体结合等方面.筛选出的9个miRNA与靶基因具有潜在调控关系,筛选出的黄芩、人参、郁金等中药具有多靶点治疗UC的潜能.结论 UC免疫机制复杂,是多因素、多靶点、多通路协同作用的结果.本研究通过对UC免疫关键基因分析,有利于探究UC发病机制,发掘潜在作用靶点及临床价值,为基础研究及有效治疗方案的完善提供新的视角.
Analysis of Key Immune Gene Mechanism and Screening of Traditional Chinese Medicine in Ulcerative Colitis
Objective Based on bioinformatics technology,the key immune target genes of ulcerative colitis(UC)were screened,and the related mechanism was analyzed.The effective traditional Chinese medicine(TCM)was screened,so as to provide theoretical support for basic research of UC and precise TCM treatment based on syndrome differentiation.Methods Key immune target genes against UC were screened by the GEO dataset,ImmPort database,SRTING database,cytoHubba plug-in,etc.The target genes were subjected to logistic univariate analysis,correlation analysis by applying the cor function,immune infiltration analysis by applying the CIBERSORT algorithm,and KEGG and GO enrichment analysis by applying the Enrichr database.miRNA was screened by searching miRWalk,RNA22,RNAInter,and TargetScan data-bases,and potential TCM was screened by the Coremine Medical database.Results Fifteen target genes with upregulated expression were screened,all of which were independent risk factors for UC.Immune infiltration analysis showed that CD4+memory-activated T cells,M0 macrophages,Ml macrophages,and neutrophils are differentially expressed between groups,and the target genes were closely associated with immune cells.KEGG analysis showed that the IL-17 signaling pathway and TNF signaling pathway were important pathways,and GO analysis indicated that biological processes mainly occurred in neutrophil chemotaxis and granulocyte chemotaxis.The cell components mainly consisted of a tertiary granule lumen and an extracellular matrix containing collagen,and the molecular functions were mainly manifested in the aspects of chemokine activity and chemokine receptor binding.The nine screened miRNAs had potential regulatory relationships with target genes,and the screened TCM such as Scutellaria baicalensis,Radix Ginseng,and Radix Curcumae had the potential for multi-target treatment of UC.Conclusions The immune mechanism of UC is complex and is the result of synergistic effects of multiple factors,targets,and pathways.By analyzing the key immune genes against UC,this study is helpful to explore the pathogenesis of UC,discover the potential targets and clinical value,and provide new perspectives for basic research and improvement of effective treatment protocols.
