Transcriptome Characteristics of Exosomes in Patients with Coronary Heart Disease Complicated with Persistent Atrial Fibrillation and Traditional Chinese Medicine Syn-dromes
Objective To investigate the unique exosomal RNA expression profiles in coronary heart disease(CHD)patients with persistent atrial fibrillation(PeAF)and different traditional Chinese medicine(TCM)syndromes,to uncover the biological basis of patients with qi and yin deficiency as well as combined phlegm and stasis,and those with blood stasis and water retention,and elucidate the internal mechanisms of these syndromes.Methods Plasma exosomes from CHD PeAF patients with qi and yin deficiency as well as combined phlegm and stasis,liver and kidney deficiency,yin deficiency with hyperactive fire,and blood stasis and and water retention were isolated for whole-transcriptome sequen-cing.Five patients were selected for each TCM syndrome,along with eight healthy volunteers as control.Bioinformatics was applied to perform biological process and KEGG pathway enrichment analyses on the unique differentially-expressed genes from patients with qi and yin deficiency as well as combined phlegm and stasis,and those with blood stasis and water retention.Results High-throughput sequencing identified 13 unique differentially-expressed microRNAs,6 mRNAs,and 3 lncRNAs in patients with qi and yin deficiency as well as combined phlegm and stasis;while 25 unique differentially-expressed microRNAs,16 mRNAs,and 5 lncRNAs were identified in patients with blood stasis and water retention.The uniquely expressed genes of the former were mainly enriched in biological processes such as hormone response,cell adhe-sion,and pathways related to lipid and atherosclerosis.In contrast,the genes of the latter were enriched in processes inclu-ding the regulation of autophagy,metabolism of fatty acid derivatives,and lipid transport,as well as the PPAR signaling pathway.Conclusion Patients with qi and yin deficiency as well as combined phlegm and stasis may exhibit pathophysio-logical states involving endocrine and immune dysfunction,as well as atherosclerosis.Meanwhile,those with blood stasis and water retention may experience abnormalities in autophagy and lipid metabolism.By examining the transcriptome profiles of exosomes,this study elucidated the internal mechanisms of TCM syndromes,providing a foundation for clinical treatment.
