Medication Rule of Traditional Chinese Medicine Prescriptions for Treating Osteo-porosis Based on Network Pharmacology
Objective To analyze the medication rule of nationally patented traditional Chinese medicine(TCM)prescriptions for treating osteoporosis and perform network pharmacology analysis.Methods TCM prescriptions for trea-ting chronic osteoporosis were retrieved from the website of the China National Intellectual Property Administration.After standardizing the names of medicines,the frequency of TCM,their tropism of tastes and natures,prescription rules,and fre-quency of drug targets were analyzed,and k-means dimensionality reduction analysis of protein-protein interaction(PPI),gene ontology(GO)-backpropagation(BP)biological function enrichment analysis,and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were conducted.Results A total of 112 patented TCM prescriptions invol-ving 156 medicinal ingredients were included in the study,with epimedium being the most frequently used herb.The nature of the medicine was commonly warm,mild,or cold;the tastes were predominantly sweet,bitter,or spicy;and they mainly acted on the liver,kidney,and spleen channels.Association rule analysis indicated common drug pair combinations such as Concha Ostreae-epimedium,Notoginseng Radix-Radix Angelicae Sinensis,and Radix Astragali-epimedium.The core targets primarily included AR,ESR1,ESR2,NR1I2,SHBG,VDR,NR3C1,GABRA family,and GABRG family.The k-means clustering analysis of the PPI network could be reduced to four categories,closely related to clinical subtypes.GO and KEGG pathway analyses demonstrated that TCM compound prescription mainly exerted its effects on the prevention and treatment of osteoporosis through neuroactive ligand-receptor interactions,chloride transmembrane transport,steroid hor-mone biosynthesis,carbohydrate digestion,and signal pathways related to substance addiction,as well as functions such as digestion and absorption,neural signaling and neurotransmission,steroid metabolism,and chloride ion transport.Conclusion The results indicate that TCM treatment for primary osteoporosis follows certain rules,providing a theoretical basis for the treatment of osteoporosis with TCM and offering new perspectives for clinical research and basic research on TCM.
