Effects of Mianan Fang on Serum Expression of IL-6,IL-1β,TNF-α and Hipp-ocampal Tissue BDNF Protein in Rats with Depression and Sleep Disorders
Objective To observe the effects of Mianan Fang on serum expression levels of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-alpha(TNF-α),and brain-derived neurotrophic factor(BDNF)in rats with depression-related sleep disorders.Methods A total of 48 specific pathogen-free(SPF)healthy female rats were divided into a model group,a Mianan Fang-treated group,a Deanxit-treated group,and a blank group using a ran-dom number table.Except for the blank group,the other groups were treated with isolation and chronic unpredictable mild stress to accelerate rapid eye movement sleep deprivation to establish the sleep deprivation depression(SDD)rat model.Be-havioral changes of rats in each group were detected by body weight,sucrose preference test,and open field test to deter-mine whether the SDD model was successfully replicated and to verify the therapeutic effect of Mianan Fang.Enzyme-linked immunosorbent assay(ELISA)was used to measure serum levels of IL-6,IL-1β,and TNF-α in rats in each group,while Western blot(WB)was used to detect the expression of BDNF protein in the hippocampus of rats.Results Behavioral measurement:Before modeling,there were no differences in body weight,sucrose preference rate,and open field test scores among the groups(P>0.05).After modeling and administration,compared with the blank group,the body weight,sucrose preference rate,and open field test scores of rats in the model group,Mianan Fang group,and Deanxit group all significantly decreased(P<0.05);compared with the model group,the body weight,sucrose preference rate,and open field test scores of rats in the Mianan Fang group and Deanxit group all increased significantly(P<0.05).ELISA results:Compared with the blank group,the serum level of IL-6,IL-1 β and TNF-α in rats in the model group increased signif-icantly(P<0.01);compared with the model group,the levels of IL-6,IL-1β and TNF-α in Mianan Fang group and the Deanxit group decreased significantly(P<0.01);there was no difference in the levels of IL-6 and IL-1β between Deanxit group and Mianan Fang group,but the level of TNF-α in the Deanxit group was lower than Mianan Fang group(P<0.05).WB results:Compared with the blank group,the expression of BDNF protein in the hippocampus of rats in the model group decreased(P<0.05);compared with the model group,the expression of BDNF protein in the Mianan Fang group and the Deanxit group increased(P<0.05);compared with the Mianan Fang group,the expression of BDNF protein in Deanxit group increased(P<0.05).Conclusion Mianan Fang can improve depressive behavior and sleep disorders in SDD model rats,and its mechanism of action may be related to reducing the levels of serum IL-6,IL-1 β,TNF-α and up-regulating the expression levels of BDNF protein in the hippocampus.
