Effect of Huanglian Nano-gelatin Paste on Expression of Inflammatory Factors and CD68 in Serum of Rats with Anal Fistula Wound
Objective To observe the effect of Huanglian nano-gelatin paste on wound healing in a rat model of anal fistula by investigating its impact on serum inflammatory factors and CD68 expression.Methods Forty clean-grade male SD rats were randomly divided into a blank group(no treatment),a model group(treated with external application of vaseline after modeling),a positive control group(treated with external application of Kangfuxin solution after modeling),and an observation group(treated with external application of Huanglian nano-gelatin paste after modeling).On the 3rd,7th,and 14th days after treatment,the formation of granulation tissue and wound healing were observed,and serum levels of interleukin-6(IL-6),interleukin-1β(IL-1β),and CD68 in granulation tissue were measured.Results The obser-vation group exhibited faster generation of fresh granulation tissue,with more abundant fibroblasts and newly formed capil-laries than the model group and the positive control group.On the 3rd,7th,and 14th days post-treatment,the wound heal-ing rate in the observation group was higher than in the positive control group.Serum levels of IL-6 and IL-1β in the ob-servation group were significantly lower than those in the model and positive control groups(P<0.05).The expression lev-el of the macrophage marker CD68 in the observation group was significantly higher than in the model and positive control groups,showing an increasing trend over time,with statistically significant differences both between groups and within groups(P<0.05).Conclusion Huanglian nano-gelatin paste can promote wound healing by modulating the inflamma-tory response and regulating CD68 expression through the action of macrophages,thereby accelerating the wound's progres-sion to the granulation proliferation and remodeling phase.
