基于网络药理学和动物实验初步探讨消瘰丸联合党参治疗甲状腺肿的作用机制

目的 基于动物实验和网络药理学初步探讨消瘰丸加党参治疗甲状腺肿的作用机制.方法 (1)通过 TCMSP数据库筛选出消瘰丸加党参的活性成分及其可能的作用靶点;通过GeneCards数据库检索甲状腺肿(Goiter,GR)疾病的相关靶点;将上述靶点导入Venny 2.1.0中对消瘰丸加党参活性成分的作用靶点与GR的疾病靶点取交集,获得消瘰丸加党参治疗GR的交集靶点;采用Cytoscape 3.8.0构建消瘰丸加党参治疗GR的"消瘰丸加党参-活性成分-交集靶点"网络,分析其关键活性成分;将交集靶点导入STRING数据库,构建蛋白-蛋白互作网络图,筛选出核心靶点;借助R包对其进行GO功能及KEGG富集分析,最后采用PyMol软件展示最佳活性成分与所有靶点之间的结合构像.(2)通过动物实验验证预测结果.购买34只6~8周龄雄性SPF级Wistar大鼠,其中8只作为正常组,其余大鼠灌服丙基硫氧嘧啶(Propylthiouracil,PTU)溶液诱导GR大鼠模型,造模成功后随机分为模型组、阳性药组、消瘰丸组、党参组,每组各6只.正常对照组坚持每日进行普通喂养并给予等量蒸馏水灌胃,模型组完成造模后同样给予等量蒸馏水灌胃,阳性药组给予优甲乐溶液灌服,消瘰丸组给予消瘰丸溶液灌服,消瘰丸加党参组给予消瘰丸加党参灌服.干预6周后采用异氟烷吸入麻醉后处死,观察大鼠病理组织、酶联免疫吸附测定法(ELISA)检测三碘甲腺原氨酸(T3)、四碘甲腺原氨酸(T4)、促甲状腺素(Thyroid stimulating hormone,TSH)水平、蛋白免疫印迹法(Western blot,WB)检测磷脂酰肌醇3-激酶(Phosphoinositide 3-kinase,PI3K)、蛋白激酶B(Protein kinase B,Akt)、B细胞淋巴瘤-2(B-cell lymphoma-2,Bcl-2)蛋白表达量.结果 (1)筛选出消瘰丸加党参相关有效成分42个,消瘰丸加党参-GR交集靶点96个;KEGG富集分析显示涉及消瘰丸加党参可能通过Akt/PI3K信号通路、细胞凋亡等发挥作用.(2)动物实验数据表明,阳性药组、消瘰丸组、党参组大鼠甲状腺组织滤泡肿胀减少,周围增生减少,间质充血较少,内部乳头状结构减少,其中以党参组甲状腺滤泡、甲状腺上皮细胞恢复最为明显.与正常组比较,模型组TSH水平、PI3K、Akt、Bcl-2蛋白表达量明显升高,差异有统计学意义(P<0.05);与模型组比较,党参组TSH水平、PI3K、Bcl-2蛋白表达量明显降低,差异有统计学意义(P<0.05),T3、T4激素水平升高.结论 消瘰丸联合党参可通过作用Bel-2等靶点、参与PI3K/Akt信号通路,发挥治疗GR的作用.
The Mechanism of Xiaoluo Pills Combined with Codonopsis Radix in the Treatment of Goiter Based on Network Pharmacology and Animal Experiments
Objective To explore the mechanism of Xiaoluo pills combined with Codonopsis Radix in the treat-ment of goiter based on animal experiments and network pharmacology.Methods(1)Active ingredients and potential tar-gets of Xiaoluo pills combined with Codonopsis Radix were screened through the TCMSP database,while goiter(GR)-rel-evant targets were retrieved from the GeneCards database.The intersecting targets between the active ingredients and GR disease targets were identified using Venny 2.1.0,resulting in the intersecting targets for Xiaoluo pills combined with Codonopsis Radix in treating GR.A"Xiaoluo pills combined with Codonopsis Radix-active ingredients-intersecting tar-gets"network was constructed using Cytoscape 3.8.0 to analyze key active ingredients.The intersecting targets were impor-ted into the STRING database to build a protein-protein interactions(PPI)network,from which core targets were screened.GO function and KEGG enrichment analyses were performed using the R package,and PyMol software was used to show the binding conformations between the best active ingredients and all the targets.(2)The predicted results were verified through animal experiments.Thirty-four male Wistar rats(6-8 weeks old,SPF-grade)were purchased,with 8 rats ser-ving as the normal group.The remaining rats were administered with propylthiouracil(PTU)solution to induce the GR rat model.After successful modeling,the rats were randomly divided into the model group,positive drug group,Xiaoluo pills group,Xiaoluo pills combined with Codonopsis Radix group,with 6 rats in each group.The normal group was given daily gavage with an equal amount of distilled water.After modeling,the model group was given distilled water by gavage;the positive drug group was given Euthyrox solution by gavage;the Xiaoluo pills group was given Xiaoluo pills solution by ga-vage;Xiaoluo pills combined with Codonopsis Radix was given both Xiaoluo pills and Codonopsis Radix by gavage.After 6 weeks of intervention,the rats were sacrificed following isoflurane anesthesia.Pathological tissue observations were conduc-ted,and enzyme-linked immunosorbent assay(ELISA)was used to measure levels of triiodothyronine(T3),tetraiodothyro-nine(T4),and thyroid-stimulating hormone(TSH).Western blot(WB)analysis was performed to detect the protein ex-pression of phosphoinositide 3-kinase(PI3 K),protein kinase B(Akt),and B-cell lymphoma-2(Bel-2).Results(1)42 active ingredients and 96 intersecting targets were identified for Xiaoluo pills combined with Codonopsis Radix in treating GR.KEGG enrichment analysis showed that Xiaoluo pills combined with Codonopsis Radix may exert effects through the Akt/PI3K signaling pathway and apoptosis.(2)Animal experiment data showed that in the positive drug group,Xiaoluo pills group,and Xiaoluo pills combined with Codonopsis Radix group,thyroid follicular swelling,surrounding hyperplasia,stromal congestion,and the internal papillary structures were reduced,with the most significant thyroid follicu-lar recovery and epithelial cell restoration observed in the Codonopsis Radix group.Compared with the normal group,TSH levels,PI3 K,Akt,and Bel-2 protein expression in the model group,were significantly elevated,with a statistically signifi-cant difference(P<0.05).Compared to the model group,TSH levels,PI3K,and Bel-2 protein expression in the Codo-nopsis Radix group were significantly reduced,with statistically significant differences(P<0.05);T3 and T4 hormone lev-els in the Codonopsis Radix group were increased.Conclusion Xiaoluo pills combined with Codonopsis Radix can exert therapeutic effects on GR by targeting Bcl-2 and participating in the PI3K/Akt signaling pathway.
