目的:本实验旨在探讨miR-26a在神经母细胞瘤中的作用及其作用机制。方法 CCK-8与EdU检测肿瘤细胞的增殖,双荧光素酶基因报告实验检测miR-26a能否与CDK8 mRNA的3’UTR区结合,qRT-PCR和western blot检测RNA及蛋白表达变化。结果SH-SY5Y细胞过表达miR-26a后细胞增殖能力明显下降,而下调miR-26a的表达细胞增殖能力显著提高;过表达miR-26a后CDK8在mRNA与蛋白水平表达降低,沉默miR-26a后CDK8在mRNA与蛋白水平表达上调,双荧光素酶基因报告实验证明miR-26a与CDK8 mRNA的3’UTR区直接结合;CDK8可以逆转miR-26a对SH-SY5Y细胞增殖的抑制作用。结论 miR-26a通过抑制CDK8的表达抑制神经母细胞瘤的增殖。
Abstract
Objective The aim of the study was to investigate the function of miR-26a in neuroblastoma cells and its mechanism. Methods CCK-8 and EdU assays were used to assess the proliferation of cells. RNA and Protein levels were detected using qRT-PCR and western blot assays. The interaction between miR-26a and CDK8 was studied using luciferase assay.ResultsThe proliferation ability of SH-SY5Y cells was enhanced after miR-26a overexpression. Whereas the proliferation ability of SH-SY5Y cells was reduced after miR-26a silenced. CDK8 was signiifcantly decreased or increased in miR-26a overexpression or knockdown cells, and luciferase assays demonstrated that miR-26a can bind to CDK8 3’UTR. Finally, the function of miR-26a can reverse by CDK8.Conclusions miR-26a inhibits neuroblastoma cell proliferation by down-regulating the expression of CDK8.
