Objective:To predict the potential mechanism of Chuanxiong Rhizoma-Salviae Miltiorrhizae Radix et Rhizoma in the treatment of spinal cord injury(SCI)based on network pharmacology.Methods:The effective components,core targets,and signa-ling pathways of Chuanxiong Rhizoma-Salviae Miltiorrhizae Radix et Rhizoma in treating SCI were obtained by network pharmacolo-gy.The rat model of SCI was established by the modified Allen method.The levels of reactive oxygen species(ROS)and malondial-dehyde(MDA)in the rats were determined by enzyme-linked immunosorbent assay(ELISA).The protein levels of phosphatidyli-nositol 3-kinase(PI3K),p-PI3K,protein kinase B(AKT),and p-AKT were determined by Western blotting.Results:The com-pound-target network consisted of 154 compounds and 484 corresponding targets.The gene ontology(GO)annotation predicted 447 GO terms.The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis showed that the key targets were involved in 126 signaling pathways.The animal experiments showed that compared with the sham group,the modeling elevated the levels of ROS and MDA(P<0.01)and down-regulated the protein levels of p-PI3K and p-AKT.Compared with the model group,high-dose Chinese medicine lowered the levels of ROS and MDA(P<0.01)and up-regulated the protein levels of p-PI3K and p-AKT(P<0.01).Conclusion:Chuanxiong Rhizoma-Salviae Miltiorrhizae Radix et Rhizoma can treat SCI in a multi-component,multi-target,and multi-pathway manner by regulating the PI3K/AKT signaling pathway.
