目的:研究夏桑菊颗粒对人冠状病毒-229E(HCoV-229E)的抑制作用.方法:采用细胞病变(CPE)法检测夏桑菊颗粒对A549细胞的安全剂量,采用CPE法检测夏桑菊颗粒对HCoV-229E引起A549细胞病变的影响,采用反转录聚合酶链式反应(RT-PCR)法检测夏桑菊颗粒对HCoV-229E于A549细胞中的复制作用;采用噻唑蓝(MTT)法检测夏桑菊颗粒对Huh-7细胞的安全剂量;采用RT-PCR法检测夏桑菊颗粒对HCoV-229E感染引起Huh-7细胞炎症介质表达的影响.结果:夏桑菊颗粒对于HCoV-229E感染A549细胞引起CPE的半数抑制浓度为23.78 mg生药/mL,选择指数为2.0,夏桑菊颗粒能够剂量依赖性地抑制A549细胞内HCoV-229E病毒载量,夏桑菊颗粒可以抑制HCoV-229E诱导Huh7细胞内炎症介质单核细胞趋化蛋白-1(MCP-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和白细胞介素-8(IL-8)的mRNA过度表达.结论:夏桑菊颗粒具有明显的抗HCoV-229E体外感染作用.
Objective:To study the inhibitory effect of Xiasangju Granules on human coronavirus-229E(HCoV-229E).Methods:The safe dose of Xiasangju Granules on A549 cells was detected by the cytopathic effect(CPE)method,and the effect of Xiasangju Granules on cytopathic effect of A549 cells caused by HCoV-229E was detected by the CPE method.Reverse transcription polymer-ase chain reaction(RT-PCR)method was used to detect the effects of Xiasangju Granules on the replication of HCoV-229E in A549 cells.The safe dose of Xiasangju Granules on Huh-7 cells was detected by Thiazolyl Blue(MTT)method,and RT-PCR meth-od was used to detect the effects of Xiasangju Granules on the expression of inflammatory mediators in Huh-7 cells caused by HCoV-229E.Results:The median inhibitory concentration of Xiasangju Granules on CPE of A549 cells infected by HCoV-229E is 23.78 mg crude drug/mL,and the selection index is 2.0.Xiasangju Granules can dose-dependently inhibit the viral load of HCoV-229E in A549 cells and inhibit the mRNA overexpression of monocyte chemoattractant protein-1(MCP-1),tumor necrosis factor-αe(TNF-α),interleukin-6(IL-6),and interleukin-8(IL-8)induced by HCoV-229E in Huh-7 cells Conclusion:Xiasangju Granules have obvious anti-infection effects of HCoV-229E in vitro.
