The Effect of"XinTong-SanLiangSan"Formula on Myocardial Ischemia Induced by Isoproterenol in Zebrafish
Objective:To study the preventive and therapeutic effect of"XinTong-SanLiangSan"Formula on myocardial ischemia induced by isoproterenol in zebrafish.Methods:The concentrations of 9.4,18.75,37.5,75,150 and 300 μL/mL of the"XinTong-SanLiangSan"Formula were used to intervene wild-type AB strain zebrafish 2 to 3 days after fertilization.The maximum tolerance concentration of the"XinTong-SanLiangSan"Formula was determined according to the toxicity phenotype and mortality of ze-brafish,thereby screening out the dosage for this study.In this experiment,the high,medium and low dose groups of"XinTong-San-LiangSan"Formula were set up,the concentrations were 75,25 and 8.3 μL/mL,respectively,the concentration of the positive con-trol Danshen dripping pills group was 500 μg/mL,the normal control group and the model control group.30 fish in each group were placed in a six-well plate,each group was placed in an incubator at 28 ℃ for 4 h,and then induced with isoproterenol hydrochloride(50 mg/mL)for 60 min to establish the myocardial ischemia model of zebrafish.After the treatment,the fluorescence intensity of apoptotic cells in the heart of zebrafish was analyzed by acridine orange staining.The zebrafish blood flow video was recorded by the heartbeat blood flow analysis software to analyze the cardiac output and blood flow velocity of zebrafish.Results:Compared with the model control group,the fluorescence intensities of apoptotic cells in the heart of zebrafish in the medium and high dose groups of"XinTong-SanLiangSan"Formula group were significantly decreased(P<0.05,P<0.01),and the improvement rates of apoptosis were 34%and 54%,respectively.The cardiac output and blood flow velocity of zebrafish in low,medium and high dose groups of"XinTong-SanLiangSan"Formula group were significantly increased(P<0.01),and the improvement rates were 52%,93%,62%and 29%,53%,32%,respectively.Conclusion:"XinTong-SanLiangSan"Formula can significantly improve myocardial ischemia induced by isoproterenol in zebrafish,which is related to improving myocardial cell apoptosis.
