Effect of Modified Shaogan Fuzi Decoction on Inflammatory Factors and Muscle Protein in CIA Rats with Sarcopenia
Objective:To explore the mechanism of modified Shaogan Fuzi Decoction on skeletal muscle damage in rheumatoid ar-thritis.Methods:A total of 36 specific pathogen-free(SPF)female Wistar rats were randomly divided into a normal group,model group,control group,and low,medium,and high dose groups of traditional Chinese medicine(TCM),with six rats in each group.Except for the normal group,other groups were used to replicate the cold syndrome model of collagen-induced arthritis(CIA)and administered with the corresponding drugs.At the end of the administration,rat serum was collected,and the content of tumor nec-rosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),and interleukin-15(IL-15)was detected by enzyme-linked immunosorbent assay(ELISA).The gastrocnemius muscle was taken,and the wet weight,gastrocnemius muscle-specific ring finger protein1(MuRF1),and muscle dystrophinF-box(MAFbx)expression were measured.Results:The wet weight,mass index,and fi-ber cross-sectional area of the gastrocnemius of muscle rats in the model group were significantly lower than those in the normal group,and the wet weight,mass index,and fiber cross-sectional area of the gastrocnemius muscle of rats in the medium and high dose groups of modified Shaogan Fuzi Decoction were significantly increased compared with those in the model group.In addition,the fiber cross-sectional area of the gastrocnemius muscle in the high dose group of TCM was significantly different from that in the control group(P<0.05).Compared with that in the normal group,the serum content of TNF-α,IL-6,IL-1β,and IL-15 of the rats in the model group was significantly increased,and the expressions of MAFbx and MuRF1 in the gastrocnemius muscle were signifi-cantly increased(P<0.05).Compared with that in the model group,the serum content of TNF-α,IL-6,IL-1β,and IL-15 of the rats in the high dose group of TCM and the control group,as well as the expressions of MAFbx and MuRF1 in the gastrocnemius muscle was decreased significantly.There was no statistically significant difference between the TCM group and the control group(P>0.05).Conclusion:Modified Shaogan Fuzi Decoction can reduce skeletal muscle damage in rheumatoid arthritis,and its possible mechanism is to reduce the content of TNF-α,IL-6,IL-15,and IL-1β inflammatory factors and lower the expression of MuRF1 and MAFbx.
