Effect of Yangjing Zhongyu Decoction on IL-1β and NF-κB in Rats with Thin Endometrium
Objective:To investigate the effect of Yangjing Zhongyu Decoction on inflammatory factors in rats with thin endometri-um.Methods:Twenty-four specific pathogen free(SPF)-grade SD female rats with regular estrous cycles were randomly divided in-to a blank group,a Chinese medicine group,and a model group,with 8 rats in each group.Rats in the Chinese medicine group and the model group were injected with 95%anhydrous ethanol into the uterine cavity to establish the rat model of thin endometrium,while the blank group received no intervention.After modeling,rats in the Chinese medicine group were administered Yangjing Zhongyu Decoction by gavage,while those in the blank and model groups were given distilled water.After three estrous cycles of treatment,uterine tissue and arterial blood were collected.Uterine endometrial thickness was measured,and hematoxylin-eosin(HE)staining was performed to observe the morphology of the uterine endometrial tissue.Serum levels of interleukin-1β(IL-1β)and nuclear factor κB(NF-κB)were measured using enzyme-linked immunosorbent assay(ELISA),and relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue were determined using quantitative PCR.Results:Compared with the blank group,the model group showed significantly reduced uterine wet weight,uterine organ index,endometrial thickness,and gland num-ber(P<0.01),significantly increased serum levels of IL-1β and NF-κB,and increased relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue(P<0.01).Compared with the model group,the Chinese medicine group exhibited significantly increased uterine wet weight,uterine organ index,endometrial thickness,and gland number(P<0.01),significantly reduced serum levels of IL-1β and NF-κB,and reduced relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue(P<0.05).Conclusion:Yangjing Zhongyu Decoction can increase endometrial thickness in rats with thin endometrium.Its mechanism of action may be related to the inhibition of NF-κB and IL-1β,which reduces inflammatory mediator levels and promotes endometrial cell proliferation.
