电针联合五子衍宗丸通过调节BDNF/TrkB/CREB信号通路对帕金森病小鼠的治疗作用

目的:观察电针联合五子衍宗丸对帕金森病小鼠黑质脑源性神经营养因子(BDNF)/酪氨酸激酶受体B(TrkB)/环磷酸腺苷反应原件结合蛋白(CREB)通路的影响.方法:将108只雄性C57BL/6小鼠按照随机数字表法分为正常组、模型组、电针组、五子衍宗丸组、联合观察组、抑制剂组、电针+抑制剂组、五子衍宗丸+抑制剂组、联合+抑制剂组9组,每组12只.除正常组、抑制剂组外,其余组给予1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)腹腔注射造模,第1天15 mg/kg,第2天20 mg/kg,第3~7天30 mg/kg,均0.2 mL/次,1次/d.自造模第1天进行干预,电针观察组别给予电针干预,五子衍宗丸观察组别早晚给予五子衍宗丸药液,抑制剂注射组别腹腔注射TrkB抑制剂ANA-12,连续干预14 d.干预结束后对比各组步态实验、旷场实验、爬杆实验结果,小鼠黑质酪氨酸羟化酶(TH)及BDNF、TrkB、CREB表达.结果:与正常组比较,模型组出现明显的运动障碍,黑质TH平均荧光强度及TH、BDNF、TrkB、CREB蛋白表达低(P<0.05);与模型组比较,电针组、五子衍宗丸组、联合观察组运动障碍均得到改善,黑质TH平均荧光强度及TH、BDNF、TrkB、CREB蛋白表达高(P<0.05);与电针组、五子衍宗丸组比较,联合观察组的运动障碍得到改善,其他指标表达增高(P<0.05).结论:电针联合五子衍宗丸可能通过调控BDNF/TrkB/CREB信号通路来改善PD小鼠的运动障碍,保护多巴胺能神经元.
Therapeutic Effect of Electroacupuncture Combined with Wuzi Yanzong Pills on Mice with Parkinson's Disease by Regulating BDNF/TrkB/CREB Signaling Pathway
Objective:To observe the effect of electroacupuncture combined with Wuzi Yanzong Pills on the BDNF/TrkB/CREB signaling pathway in mice with Parkinson's disease(PD).Methods:A total of 108 male C57BL/6 were randomly divided into nine groups,including the normal group,model group,electroacupuncture group,Wuzi Yanzong Pill group,combined treatment group,in-hibitor group,electroacupuncture+inhibitor group,Wuzi Yanzong Pill+inhibitor group,and combined treatment+inhibitor group,with 12 mice in each group.Except for the normal group and the inhibitor group,the other groups were given 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)by intraperitoneal injection for modeling,with 15 mg/kg on the first day,20 mg/kg on the second day,and 30 mg/kg on the third to seventh days,0.2 mL each time,once a day.The intervention was performed on the first day of modeling.The electroacupuncture group was treated with electroacupuncture intervention,and the Wuzi Yanzong Pill group was fed with Wuzi Yanzong Pill solution in the morning and evening for 14 consecutive days.The inhibitor group was intraperitoneally injec-ted with ANA-12,a TrkB inhibitor for 14 consecutive days.After the intervention,the results of the gait experiment,open field ex-periment,and pole climbing experiment were compared,as well as the expressions of tyrosine hydroxylase(TH),BDNF,TrkB,and CREB in substantia nigra.Results:Compared with the normal group,the model group showed obvious dyskinesia,and the average fluorescence intensity of TH in substantia nigra and the protein expression of TH,BDNF,TrkB,and CREB decreased significantly(P<0.05).Compared with that of the model group,the dyskinesia of the electroacupuncture group,Wuzi Yanzong Pill group,and combined treatment group was improved,and the average fluorescence intensity of TH in substantia nigra and the protein expression of TH,BDNF,TrkB,and CREB were increased(P>0.05),while the dyskinesia of combined treatment group was improved com-pared with the electroacupuncture group and Wuzi Yanzong Pill group.The expression of other indexes increased(P<0.05).Con-clusion:Electroacupuncture combined with Wuzi Yanzong Pills may improve the dyskinesia of mice with PD and protect dopaminer-gic neurons by regulating the BDNF/TrkB/CREB signaling pathway.
