摘要

目的:观察阴虚胃痛胶囊对慢性萎缩性胃炎(CAG)大鼠胃黏膜超微结构及炎症介质的影响.方法:将72只无特定病原体(SPF)级斯泼累格·多雷(SD)大鼠分为正常组13只和造模组59只,造模组给予N-甲基-N'-硝基-N-亚硝基胍自由饮+灌胃,同时给予饥饱失常法复合因素干预制备CAG大鼠模型.将成模大鼠按随机数字表法分为模型组、维酶素组及阴虚胃痛胶囊低剂量、中剂量、高剂量组.阴虚胃痛胶囊低剂量、中剂量、高剂量组分别给予205.2 mg/kg、410.4 mg/kg、820.8 mg/kg阴虚胃痛胶囊溶液灌胃,维酶素组给予270 mg/kg维酶素溶液灌胃,正常组与模型组给予等剂量生理盐水灌胃,均1次/d,共干预8周.苏木精-伊红(HE)染色观察大鼠胃黏膜病理情况;透射电镜观察大鼠胃黏膜超微结构变化;实时荧光定量聚合酶链反应检测胃组织中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的基因表达;蛋白质免疫法检测胃组织中炎症介质TNF-α、IL-1β和抗炎因子白细胞介素-4(IL-4)、白细胞介素-10(IL-10)的蛋白表达.结果:与正常组比较,模型组HE染色显示腺体减少;透射电镜显示壁细胞和主细胞结构被明显破坏,TNF-α、IL-1β的表达高(P<0.05),IL-4、IL-10表达低(P<0.05).与模型组比较,阴虚胃痛胶囊中剂量、高剂量组及维酶素组HE提示腺体改善,透射电镜提示壁细胞和主细胞结构恢复,胃黏膜中TNF-α、IL-1β表达低(P<0.05),IL-4、IL-10表达高(P<0.05).结论:阴虚胃痛胶囊可明显促进CAG大鼠胃黏膜异常超微结构的恢复,抑制炎症介质TNF-α、1L-1β表达,促进抗炎因子IL-4、IL-10表达.

Abstract

Objective:To observe the effects of Yinxu Weitong Capsules on the ultrastructure of gastric mucosa and inflammatory mediators in rats with chronic atrophic gastritis(CAG).Methods:A total of 72 specific pathogen-free(SPF)Sprague-Dawley(SD)rats were divided into a normal group(n=13)and an experimental group(n=59).The experimental group was given N-methyl-N'-nitro-N-nitrosoguanidine in drinking water and intragastric administration,combined with irregular feeding to induce a CAG rat model.The modeled rats were randomly divided into a model group,a vitacoenzyme group,and low-,medium-,and high-dose Yinxu Weitong Capsules groups.The Yinxu Weitong Capsules groups received intragastric administration of Yinxu Weitong Capsules at doses of 205.2 mg/kg,410.4 mg/kg,and 820.8 mg/kg,respectively.The vitacoenzyme group received vitacoenzyme solution at 270 mg/kg,and the normal and model groups received an equivalent dose of saline,all once daily for eight weeks.Hematoxylin-eo-sin(HE)staining was used to observe the pathological changes in the gastric mucosa.Transmission electron microscopy was used to observe ultrastructural changes.Real-time quantitative PCR was used to detect the gene expression of tumor necrosis factor-alpha(TNF-α)and interleukin-1 beta(IL-1β)in gastric tissue.Western blot was used to detect the protein expression of pro-inflamma-tory factors TNF-α and IL-1β and anti-inflammatory factors interleukin-4(IL-4)and interleukin-10(IL-10).Results:Compared with the normal group,HE staining in the model group showed reduced glandular tissue,and transmission electron microscopy showed significant structural damage to parietal and chief cells.Moreover,the expression of TNF-α and IL-1 β was higher(P<0.05),and the expression of IL-4and IL-10 was lower(P<0.05).Compared with the model group,the medium-and high-dose Yinxu Weitong Capsules groups and the vitacoenzyme group showed improved glandular tissue in HE staining,restored parietal and chief cell structures in transmission electron microscopy,lower expression of TNF-α and IL-1 β in gastric mucosa(P<0.05),and higher expression of IL-4 and IL-10(P<0.05).Conclusion:Yinxu Weitong Capsules significantly promote the recovery of abnor-mal gastric mucosal ultrastructure in CAG rats,inhibit the expression of pro-inflammatory factors TNF-α and IL-1 β,and promote the expression of anti-inflammatory factors IL-4 and IL-10.
