Content and Significance of Xanthopurpurin in Rubia cordifolia before and after Processing Based on HPLC
Objective:To establish a high-performance liquid chromatography(HPLC)method for comparing the changes in the content of xanthopurpurin before and after the processing of Rubia cordifolia and to explore its significance.Methods:The analysis was performed using an ACE Excel 5 C18-PFP column with a mobile phase of acetonitrile-water(60:40)at a flow rate of 1.0 mL/min.The detection wavelength was set at 245 nm,and the column temperature was maintained at 30 ℃.The processing of R.cordi-folia from different regions was entrusted to Yulin Guangjitang Pharmaceutical Technology Co.,Ltd.,Anguo Juyaotang Pharmaceu-tical Co.,Ltd.,Lanzhou Antaitang Traditional Chinese Medicine Decoction Pieces Co.,Ltd.,Hebei Renxin Pharmaceutical Co.,Ltd.,and Tongjuntang Pharmaceutical Co.,Ltd.The content of xanthopurpurin before and after processing was determined.Re-sults:The content of xanthopurpurin in raw R.cordifolia ranged from 0.040 to 0.169 mg/g.After processing into charred R.cordifo-lia at 110 ℃ to 130 ℃,the content ranged from 0.309 to 0.380 mg/g,and at 350 ℃ to 370 ℃,the content ranged from 0.513 to 1.169 mg/g.Xanthopurpurin showed good linearity in the range of 6.712 to 83.905 μg/mL,with a correlation coefficient(r)of 1.000 0.The average recovery rate was 101.3%,with a relative standard deviation(RSD)of 2.1%.Conclusion:The HPLC meth-od can effectively distinguish between raw R.cordifolia and its charred product based on the content of xanthopurpurin.
