Effects of Moxibustion on PI3K-AKT Signaling Pathway in Cancer-related Fatigue Mice
Objective:To observe the effects of moxibustion on cancer-related fatigue(CRF)in mice and its impact on the phos-phoinositide 3-kinase(PI3K)-protein kinase B(AKT)signaling pathway.Methods:BALB/C mice were randomly divided into four groups:blank control group(n=10),model group(n=15),moxibustion group(n=15),and infrared group(n=15).The blank control group received no treatment.The model group was induced with a CRF animal model by subcutaneously transplanting 4T1 breast cancer cells and administering cisplatin.The moxibustion and infrared groups received moxibustion and infrared radiation at the Guanyuan(BL 26)for 4 weeks,respectively.After the intervention,the behavior of the mice was assessed through fatigue tread-mill experiment and tail suspension test.Serum levels of interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α)were meas-ured using enzyme-linked immunosorbent assay(ELISA).Western blot was used to detect protein expression of phosphorylated PI3K(p-PI3K),AKT,phosphorylated AKT(p-AKT),B-cell lymphoma 2(Bcl-2),and caspase-3 in tumor tissues.Reverse tran-scription-polymerase chain reaction(RT-PCR)was used to detect the mRNA expression of IL-6 and PI3K in tumor tissues.Re-sults:Compared with the blank control group,the model group exhibited a reduction in fatigue treadmill time and distance,an in-crease in spleen index,treadmill shock frequency,and tail suspension time,as well as elevated serum levels of IL-6 and TNF-α.Compared with the model group,the intervention groups showed an increase in treadmill time and distance,a reduction in shock fre-quency and tail immobility time,a decrease in tumor weight,spleen index,IL-6,TNF-α,p-PI3K,p-AKT,IL-6 mRNA,and PI3K mRNA levels.In the moxibustion group,Bcl-2 levels were reduced,while caspase-3 expression was increased.Compared with the infrared group,the moxibustion group had reduced tail immobility time,IL-6,TNF-α,p-PI3K,and IL-6 mRNA expression.Conclu-sion:Moxibustion can alleviate CRF,and its mechanism may involve the PI3K-AKT signaling pathway mediated by inflammatory mediators.Infrared radiation plays a significant role in the effectiveness of moxibustion.
