目的:采用超高效液相色谱-线性离子阱-静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap/MS/MS)对丁氏三阳透解汤干浸膏及含药血清的活性成分进行鉴定,探寻丁氏三阳透解汤药效物质基础.方法:通过UPLC-Q-Orbitrap/MS/MS技术对丁氏三阳透解汤干浸膏、含药血清、空白血清进行主要成分鉴定,运用Xcalibur软件进行数据采集和分析,利用成分裂解数据库对质谱峰进行指认,确定丁氏三阳透解汤活性成分,用生物信息分析活性成分的药理作用.结果:共鉴定出丁氏三阳透解汤有效活性成分18种,包括木犀草素、4'-O-甲氧基葛根素、芦荟大黄素-8-O-葡萄糖苷、柚皮素、大黄素-8-O-β-D-葡萄糖苷、没食子酸、黄芩苷、山柰酚等.生物信息分析结果显示丁氏三阳透解汤主要活性成分可作用于炎症性疾病、消化系统疾病、心血管系统疾病等;丁氏三阳透解汤主要通过氧化磷酸化、过氧化物酶体增殖物激活受体(PPAR)炎症信号通路、类固醇激素生物合成和脂肪酸代谢等通路发挥作用.结论:成功鉴定了 18种丁氏三阳透解汤的有效活性成分,丁氏三阳透解汤具有"多成分-多靶点-多途径"的药理作用.
Component Identification and Material Basis of Dingshisanyangtoujie Based on UPLC-Q-Orbitrap/MS/MS and Bioinformatics Analysis Technology
Objective:This study aimed to identify the active components of Dingshisanyangtoujie Decoction Dry Extract and drug-containing serum by using ultra-performance liquid chromatography-linear ion trap-electrostatic field orbitrap high-resolution mass spectrometry(UPLC-Q-Orbitrap/MS/MS)and explore the material basis of pharmacodynamics of Dingshisanyangtoujie Decoction.Methods:Firstly,the main components of Dingshisanyangtoujie Decoction Dry Extract,drug-containing serum,and blank serum were identified by UPLC-Q-Orbitrap/MS/MS,and Xcalibur software was used for data acquisition and analysis.The mass spectral peaks were identified according to the component cleavage database,and the active components of Dingshisanyangtoujie Decoction were determined.The pharmacological action of the active components was clarified by bioinformatics analysis.Results:A total of 18 effective active components of Dingshisanyangtoujie Decoction were identified,including luteolin,4'-O-methoxypuerarin,aloe emodin-8-O-glucoside,naringenin,emodin-8-O-β-D-glucoside,gallic acid,baicalin,and kaempferol.The results of bioinformatics a-nalysis showed that the main active components of Dingshisanyangtoujie Decoction could act on inflammatory diseases,digestive system diseases,and cardiovascular system diseases.Dingshisanyangtoujie Decoction mainly played a role through oxidative phos-phorylation,peroxisome proliferator-activated receptor(PPAR)inflammatory signaling pathway,steroid hormone biosynthesis,and fatty acid metabolism pathways.Conclusion:Eighteen effective active components of Dingshisanyangtoujie Decoction are successful-ly identified,and Dingshisanyangtoujie Decoction has the pharmacological effect of"multiple components,multiple targets,and mul-tiple pathways".
