Inhibitory Effect of HZRZJQ Recipe on Esophageal Precancerous Lesions by Regulating Proliferation and Differentiation-related Factors
Objective:This paper aimed to observe the effect of HZRZJQ Recipe on the protein and gene expression of peptidyl prolyl cis-trans isomerase(Pin1)and mammalian target protein of rapamycin(mTOR)related to the proliferation and differentiation of esophageal precancerous lesions in mice and clarify the mechanism of HZRZJQ Recipe.Methods:A total of 130 mice were ran-domly divided into a normal control group,lesion model group,western medicine treatment group,traditional Chinese medicine(TCM)prevention group,and TCM treatment group.In addition,4-nitroquinoline-oxide(4-NQO)was used to establish the esopha-geal precancerous lesion model,and immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR)methods were used to detect the effect of HZRZJQ Recipe on the expression of Pin1,mTOR protein,and gene in esophageal tissue.Results:The esophageal precancerous lesion model was successfully established at the end of the 14th week.Compared with those in the normal control group,Pin1,mTOR protein,and gene expression levels in the lesion model group were increased significantly(P<0.05).Compared with those in the lesion model group,the expression levels in the western medicine treatment group,TCM preven-tion group,and TCM treatment group were decreased significantly(P<0.05).Compared with that in the TCM treatment group,the protein and gene expression in the TCM prevention group was decreased.Conclusion:4-NQO is an effective drug for establishing e-sophageal precancerous lesion model in mice,and HZRZJQ Recipe can inhibit the proliferation and differentiation of esophageal precancerous lesions by inhibiting the expression of Pin1,mTOR protein,and gene in mice with esophageal precancerous lesions.Preventive TCM medication can prevent esophageal cancer development.
