Optimization of extraction process and study of in vitro antioxidant activity of total flavonoids from Perilla frutescens leaves by orthogonal test
Using Perilla frutescens leaves as the research subject,based on single-factor experiments,the extraction process of total flavonoids from Perilla frutescens leaves was optimized through orthogonal experimental design to explore the antioxidant activity of total flavonoids.The yield of total flavonoids from Perilla frutescens leaves was taken as the evaluation index,and ethanol concentration,liquid-to-material ratio,extraction temperature,and extraction time were optimized.The results showed that the main and secondary influencing factors on the yield of total flavonoids from Perilla frutescens leaves were in the following order:Extraction temperature>ethanol concentration>liquid-to-material ratio>extraction time.The optimal extraction parameters for total flavonoids from Perilla frutescens leaves were found to be an ethanol.concentration of 50%,a liquid-to-material ratio of 20 mL/g,an extraction temperature of 70 ℃,and an extraction time of 45 minutes.Under these conditions,the yield of total flavonoids from Perilla frutescens leaves was 4.05%.Total flavonoids from Perilla frutescens leaves exhibited strong scavenging effects on DPPH free radicals and hydroxyl radicals,with IC50 values of 160 mg/L and 196 mg/L,respectively.The study indicates that total flavonoids from Perilla frutescens leaves possess strong antioxidant properties,making them a high-quality natural antioxidant.
