Fluorescence labeling,identification,and in vivo distribution of Millettia speciosa polysaccharide
The study was to explore the absorption and distribution of Millettia speciosa polysaccharide in mice.The Millettia speciosa polysaccharide Rhodamine B fluorescent marker(MSP-Rh B)was obtained by labeling Millettia speciosa polysaccharide with Rhodamine B.After the labels were identified by fluorescence microscope and infrared spectrum scanning,MSP-Rh B was given to mice by intragastric administration.Samples were collected at 1,3,6,12,24 h,respectively,and the distribution of MSP-Rh B in vivo and in vitro was observed by small animal in vivo imaging system.The results showed that fluorescence labeling of Millettia speciosa polysaccharide was successful.After intragastric administration,MSP-Rh B was mainly distributed in gastrointestinal tract,liver and kidney.MSP-Rh B reached each segment of the small intestine at 1 h,reached its maximum at about 3 h,and stayed in the small intestine for 24 h.Studies have shown that MSP-Rh B is absorbed through the small intestine,enters the systemic circulation first,and then distributes to the liver and kidney,and the fluorescence signal gradually decreases with time.
