Study on induction of yeast cell wall release of mannan through phase wise culture of two bacteria coupling
The experiment aimed to achieve the efficient release and oligosaccharidation of mannan from yeast cell walls.A novel dual-strain coupled phased cultivation model is proposed in the experiment to induce the release of mannan from yeast cell walls,and the effectiveness of polysaccharide release was verified by screening inducing strains and dual-strain phased fermentation.The results showed that by comparing the effects of four Bacillus strains on the cell wall rupture rate and mannan release rate of brewer's yeast,a Bacillus subtilis-FMT-K01-Saccharomyces cerevisiae dual-strain phased cultivation model was established.After 72 hours of dual-strain phased cultivation,the yeast cell wall rupture rate reached 96.63%,and the mannan release rate reached 83.87%.Through the depolymerization analysis of mannan,it was found that the dual-strain phased cultivation model achieved 37.27%oligosaccharidation of yeast mannan.The study indicates that the dual-strain coupled phased fermentation model proposed in this research can accelerate the release of mannan from yeast cell walls and the oligosaccharidation of polysaccharides,and it has certain reference significance for the preparation of other biological polysaccharides and their oligosaccharides.
