益母生化散对LPS诱导的小鼠子宫内膜炎血清激素和TLR4/NF-κB信号通路相关因子的影响

试验旨在探究益母生化散对子宫内膜炎模型小鼠血清激素水平和Toll样受体4/核转录因子-κB(TLR4/NF-κB)信号通路相关因子表达水平的影响.选取日龄、体型、体重相近的雌性小鼠40只,随机分为4组,每组10个重复,每个重复1只.对照组小鼠子宫灌注20 μL生理盐水,模型组子宫灌注25 μL脂多糖(LPS,1 g/L),药物组灌胃2.0 g/kg益母生化散,治疗组子宫灌注25μL LPS+2.0 g/kg益母生化散灌胃,连续灌胃3d,2次/d.利用放射免疫分析法检测血清中雌二醇(E2)、孕酮(Pg)、促黄体生成素(LH)和促卵泡生成素(FSH)含量,ELISA方法检测血清中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子(TNF-α)含量,RT-PCR方法检测TLR4/NF-κB信号通路相关因子Toll样受体4(TLR4)、髓样分化因子(MyD88)、核因子-κB p65(NF-κBp65)、IL-1β、IL-6、IL-8、TNF-α mRNA的表达.结果显示:与对照组相比,模型组血清E2、LH和FSH含量明显降低,而Pg含量升高,IL-1β、IL-6、IL-8和TNF-α的分泌量及其mRNA基因表达量明显升高,TLR4、MyD88、NF-κB p65 mRNA的表达明显上调;与模型组相比,治疗组血清E2、LH和FSH含量明显升高,Pg含量降低,IL-6、IL-8、IL-1β和TNF-α的分泌量及其mRNA基因表达明显降低,TLR4、MyD88、NF-κBp65 mRNA的表达量明显下调.研究表明,益母生化散通过调节激素含量和抑制TLR4/NF-κB信号通路相关因子水平的变化,对LPS诱导的小鼠子宫内膜炎起到缓解作用.
Effects of Yimu Shenghuasan on serum hormones and TLR4/NF-κB signaling pathway related factors in LPS-induced endometritis in mice
The experiment is designed to explore the impact of Yimu Shenghuasan on the serum hormone levels and the expression levels of factors related to the TLR4/NF-κB signaling pathway in a mouse model of endometritis.A total of 40 female mice,with similar age,body size,and weight,were randomly divided into four groups with 10 replicates per group and one mouse per replicate.Mice in the control group were instillated with 20 μL normal saline,the model group was instillated with 25 μL lipopolysaccharide(LPS,1 g/L),the drug group was instillated with 2.0 g/kg Yimu Shenghuasan,and the treatment group was instillated with 25 μL LPS+2.0 g/kg Yimu Shenghuasan,continuously gavage for three days,twice a day.The contentrations of estradiol(E2),progesterone(Pg),luteinizing hormone(LH),and follicle-stimulating hormone(FSH)in the serum was detected by radioimmunoassay,the contentrations of interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-8(IL-8),tumor necrosis factor-α(TNF-α)in the serum was detected by ELISA method,and the expression of factors related to the TLR4/NF-κB signaling pathway,including Toll-like receptor 4(TLR4),myeloid differentiation factor(MyD88),nuclear factor-KB p65(NF-κB p65),IL-1β,IL-6,IL-8,TNF-α mRNA was detected by RT-PCR method.The results showed that compared with the control group,the serum E2,LH,and FSH contentrations in the model group was significantly reduced,while Pg content increased,the secretion and mRNA gene expression of IL-1β,IL-6,IL-8,and TNF-α were significantly increased,and the expression of TLR4,MyD88,NF-κB p65 mRNA was significantly upregulated.Compared with the model group,the serum E2,LH,and FSH content in the treatment group was significantly increased,Pg content reduced,the secretion and mRNA gene expression of IL-6,IL-8,IL-1β,and TNF-αwere significantly reduced,and the expression of TLR4,MyD88,NF-κB p65 mRNA was significantly downregulated.The study indicates that Yimu Shenghuasan,by regulating the content of hormones and inhibiting the changes in the levels of factors related to the TLR4/NF-κB signaling pathway,has a relieving effect on LPS-induced mouse endometriosis.
