Effects of patchouli essential oil on growth performance,intestinal barrier permeability,and antioxidant capacity in a mice model
The experiment aimed to study the effects of different levels of patchouli essential oil(PEO)added to the diet on the growth performance,intestinal barrier permeability,and antioxidant capacity of Kunming mice.Thirty-six male Kunming mice of similar weight at 21 days of age were randomly divided into three groups,fed with a basic diet(AIN-93G)(control group),a basic diet+200 mg/kg PEO(ME group),and a basic diet+400 mg/kg PEO(HE group),with six replicates per group and two mice per replicate.The preliminary trial lasted for three days,and the formal experiment lasted for 28 days.The results showed that compared with the control group,the average daily weight gain(ADG)and final body weight of the Kunming mice in the ME group were significantly increased(P<0.05),and the feed-to-gain ratio(F/G)was significantly reduced(P<0.05).Compared with the control group,the villus height(VH)and the ratio of villus height to crypt depth(V/C)of the small intestine in the Kunming mice in the ME group were significantly increased(P<0.05).Compared with the control group,the levels of diamine oxidase(DAO),D-lactic acid(D-LAC),and myeloperoxidase(MPO)in the small intestine tissue of the Kunming mice in both the ME and HE groups were significantly reduced(P<0.05),while the relative expression levels of mucin(Mucin1),tight junction protein-1(ZO-1),occluding(Occludin),and tight junction protein-1(Claudin-1)mRNA in the small intestine tissue were significantly increased(P<0.05).In addition,the transmembrane resistance(TER)of intestinal mucosa of Kunming mice in the ME group was significantly improved(P<0.05),and the permeability of FITC-labeled dextran(FD4)was significantly reduced(P<0.05).Compared with the control group,the levels of DAO,D-LAC,and MPO in serum of Kunming mice in the ME group and HE group decreased significantly(P<0.05),the total antioxidant capacity(T-AOC)increased significantly(P<0.05),and the level of malondialdehyde(MDA)decreased significantly(P<0.05).The study indicates that PEO can significantly improve the growth performance,enhance the intestinal barrier function,and the antioxidant capacity of Kunming mice,with the best effect when the PEO addition is 200 mg/kg.
