Based on expression characteristics of zebrafish SLC17A8 genes,effect on melanin transport was verified
The study aimed to investigate the mechanism of nutritional metabolism of melanin formation in Lanping black-boned sheep.This experiment was based on the Homo sapiens solute carrier family 17,member 8(SLC17A8)gene screened by the whole genome resequencing of the subject group as a candidate gene,and the SLC17A8 gene was knocked out on zebrafish using the CRISPR/Cas9 system knockout technology.The results showed that compared with the wild-type zebrafish,36 h and 42 h mutant zebrafish had abnormal yolk development and individual differences in the amount of yolk melanin deposition,and it was more obvious at 36 h than at 42 h.However,the yolk melanin changes were not obvious at 72 h knockout.The study indicates that this gene has a role in the pre-melanin transporter phase,but has little effect in the later phase.
