响应面法优化黄柏多糖的提取工艺及其抗氧化作用研究

试验采用响应面法优化黄柏中多糖成分的提取工艺,并研究其抗氧化活性.选择多糖提取中影响较大的4个因素(提取温度、提取时间、超声功率和液料比)为考察对象,以单因素试验结果为基础,采用Box-Behnken设计响应面法优化黄柏多糖的提取工艺,通过测定黄柏多糖对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟基自由基清除作用,评价黄柏多糖的抗氧化活性.结果显示,响应面优化的最优提取工艺为提取温度79℃、提取时间54 min、超声功率59 W、液料比23 mL/g,此时黄柏多糖得率为达到3.19%.黄柏多糖浓度为5g/L时,对DPPH自由基和羟基自由基的最大清除率分别为61%和39%.研究表明,优化得到的黄柏多糖超声提取工艺稳定可行,黄柏多糖具有一定的抗氧化活性.
Study on optimization of extraction process from Phellodendron amurense Rupr.polysaccharide by response surface methodology and its antioxidant activity
The experiment used the response surface method to optimize the extraction process of polysaccharides from Phellodendron amurense Rupr.and studied its antioxidant activity.Four factors with significant impact on polysaccharide extraction(extraction temperature,extraction time,ultrasonic power,and liquid-to-solid ratio)were selected as the subjects of investigation.Based on the results of single-factor experiments,the Box-Behnken design response surface method was used to optimize the extraction process of Phellodendron amurense Rupr.polysaccharides.The antioxidant activity of Phellodendron amurense Rupr.polysaccharides was evaluated by measuring their scavenging effects on 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radicals and hydroxyl radicals.The results showed that the optimal extraction process obtained by the response surface optimization was an extraction temperature of 79 ℃,an extraction time of 54 minutes,an ultrasonic power of 59 W,and a liquid-to-solid ratio of 23 mL/g,at which the yield of Phellodendron amurense Rupr.reached 3.19%.At a concentration of 5 g/L,the maximum scavenging rates of DPPH and hydroxyl radicals by Phellodendron amurense Rupr.polysaccharides were 61%and 39%,respectively.The study indicates that the optimized ultrasonic extraction process of Phellodendron amurense Rupr.polysaccharides is stable and feasible,and Phellodendron amurense Rupr.polysaccharides possess certain antioxidant activity.
