Optimization of extraction method of total flavonoids from Rehmannia glutinosa and study on its antioxidant activity
The study aimed to optimize the extraction process of total flavonoids from Rehmannia glutinosa using the response surface methodology and to evaluate the total flavonoid content and antioxidant capacity in the radial striations,no-radial striations and root of Rehmannia germplasm QH-1 and Jinjiu.The results showed that the optimal extraction conditions for total flavonoids from Rehmannia glutinosa were a liquid-to-solid ratio of 30 mL/g,ethanol volume fraction of 50%,extraction temperature of 50 ℃,and extraction time of 40 min,with a total flavonoid extraction rate of 0.130 2%.The content of total flavone of Rehmannia glutinosa in both germplasm was non-radial striations parts>radial striations parts.The IC50 values for scavenging 1,l-diphenyl-2-picrylhydrazyl(DPPH)free radicals of QH-1 Rehmannia glutinosa radial striations,no-radial striations,and root were 0.016,0.009,and 0.009 g/L,respectively,while those for Jinjiu Rehmannia glutinosa were 0.006,0.009,and 0.010 g/L,respectively.The IC50 values for scavenging hydroxyl free radicals of QH-1 Rehmannia glutinosa radial striations,no-radial striations,and root were 0.054,0.035,and 0.047 g/L,respectively,while those for Jinjiu Rehmannia glutinosa were 0.024,0.032,and 0.034 g/L,respectively.The study indicates that the total flavonoids from Rehmannia glutinosa possess strong antioxidant activity,which can be used as natural and green antioxidants in animal production.
