半边风多酚提取工艺优化及其抗氧化和抗菌活性研究

为探究半边风多酚提取工艺及其抗菌和抗氧化活性,研究采用微波辅助提取法,在单因素试验的基础上通过响应面试验优化半边风多酚提取工艺,并以羟自由基和1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率为指标测定半边风多酚的抗氧化能力,以抑菌圈直径、最低抑菌浓度(MIC)、最低杀菌浓度(MBC)为指标测定其抑菌能力.结果显示:半边风多酚最优提取参数为乙醇体积分数44%、液料比60mL/g、提取时间2.7 min、提取功率266 W,多酚得率可达1.26%;与L-抗坏血酸相比,半边风多酚具有一定的羟自由基、DPPH自由基清除能力,具有抗氧化活性;半边风多酚对金黄色葡萄球菌、大肠杆菌抑菌圈直径分别为9.50 mm和8.00 mm,MIC分别为0.469 g/L和0.938 g/L,MBC分别为1.875 g/L和0.938 g/L.研究表明,半边风多酚具有抗氧化和抗菌双重活性,可以开发成饲料添加剂在动物生产中应用.
Study on optimization of extraction process of polyphenols of Plerospermum heterophyllum Hance and its antioxidant and antibacterial activities
To explore the extraction process and antibacterial and antioxidant activities of polyphenols of Plerospermum heterophyllum Hance.This study used microwave-assisted extraction method to optimize the extraction process of polyphenols of Plerospermum heterophyllum Hance based on single factor experiments through response surface methodology.The antioxidant capacity of polyphenols of Plerospermum heterophyllum Hance was measured using hydroxyl radical and DPPH radical scavenging rate as indicators,and the antibacterial capacity was measured using inhibition zone diameter,minimum inhibitory concentration(MIC),and minimum bactericidal concentration(MBC)as indicators.The results showed that the optimal extraction parameters for polyphenols of Plerospermum heterophyllum Hance were ethanol volume fraction of 44%,liquid-solid ratio of 60 mL/g,extraction time of 2.7 minutes,extraction power of 266 W,and polyphenol yield of 1.26%.Compared with L-ascorbic acid,the polyphenols of Plerospermum heterophyllum Hance have a certain hydroxyl radical and DPPH radical scavenging rate,and have antioxidant activity.The diameters of the bacteriostatic circles of polyphenols of Plerospermum heterophyllum Hance against Staphylococcus aureus and Escherichia coli were 9.50 and 8.00 mm respectively,the MIC was 0.469 and 0.938 g/L respectively,and the MBC was 1.875 and 0.938 g/L respectively.The results show that the polyphenols of Plerospermum heterophyllum Hance have both antioxidant and antibacterial activities and can be developed into feed additives for animal production.
