Optimization of cultivation conditions for Lactobacillus rhamnosus TCCC 10035
The experiment aimed to optimize the culture conditions for Lactobacillus rhamnosus TCCC 10035.Through single-factor experiments and response surface methodology,the culture conditions for Lactobacillus rhamnosus TCCC 10035 were optimized.The results showed that glucose,manganese sulfate,and vitamin C were important components of the fermentation medium for Lactobacillus rhamnosus TCCC 10035.The response surface results obtained the optimal medium parameters as glucose 25.00 g/L,manganese sulfate 0.32 g/L,and vitamin C 0.24 g/L.Further optimization of the culture conditions led to the best culture conditions:an initial pH value of 6.3,an inoculation volume of 4%,static cultivation for 22 hours,and a cultivation temperature of 37℃.Under these conditions,the number of bacteria obtained reached 5.36×109 CFU/mL,which was a 121%increase compared to before optimization.The study indicates that the optimized medium components and fermentation conditions can increase the number of viable bacteria of Lactobacillus rhamnosus TCCC 10035,which has application value in animal husbandry production.
