Analysis of population genetic polymorphism of the A-FABP gene in three varieties of chicken
The experiment aimed to investigate the population genetic polymorphism of fat adipocyte type fatty acid binding protein(A-FABP)gene among Bashang long-tail chicken,Dongxiang black chicken,and Baiyu Leghorn chicken.The SNP detection of A-FABP gene sequences in different breeds were carried out by the PCR-SSCP method.The results showed that there were four mutation sites:g.09A>G,g.110T>C,g.1607A>G,and g.1727A>C,all of which were first discovered in chickens.The mutations at the two loci of g.1607A>G and g.1727A>C occurred only in the Bashang long-tail chicken,and Bashang long-tail chickens had the highest polymorphic information content(mean PIC value 0.328 0),and the highest observed heterozygosity(average Ho 0.594 5).Hardy-Weinberg equilibrium test found that three mutation sites(g.09A>G,g.1607A>G and g.1727A>C)in Bashang long-tail chickens were in equilibrium(P>0.05),and two mutation sites(g.09A>G,g.110T>C)in Baiyu Leghorn chicken were in equilibrium.(P>0.05),and the other sites are in an unbalanced state(P<0.05).The study indicates that the genetic polymorphism of A-FABP gene in Bashang long-tailed chicken was higher than that in Dongxiang black chicken and Baiyu Leghorn chicken.
