Advances in the construction of Escherichia coli chassis cells for the synthesis of N-glycosylated proteins
N-glycosylation is one of the major post-translational modifications in nature,and its effects on protein structure and function are important.With the rapid development in the field of glycoengineering,it has become more common to accomplish N-glycosylation modifications of therapeutic proteins in Escherichia coli(E.coli).Through the use of gene editing technology,E.coligenome can be modified to gain new features and productivity,which can boost the production of target glycoproteins.Using gene editing technology to modify the E.coli genome and create E.coli chassis cells,as well as improving N-glycosylation efficiency to increase the synthesis of N-glycosylated proteins,are discussed in this study.In addition to providing a foundation for the creation of engineered strains with N-glycosylation modification,this work offers techniques for increasing the efficiency of″glycoprotein factories″and improving glycoprotein output.
