N6-甲基腺嘌呤(N6-methyladenosine,m6A)是真核生物RNA中的一种最为普遍的表观遗传修饰,拥有大量的识别蛋白以及复杂的调控网络,能够发挥基因表达调控及细胞命运决定等功能.m6A不仅在转录后水平调控基因表达,也广泛参与了基因转录水平的调控.m6A可以通过影响RNA聚合酶启动子近端暂停、R-环形成等过程对转录起直接调控作用,也能够通过影响染色质的结构及活性间接调控转录.本文对m6A及m6A的共转录写入进行了简要介绍,重点讨论了m6A对基因的转录水平调控以及相关的生物学功能.
Regulation of gene transcription by RNA m6A
N6-methyladenine(m6A)is the most common epigenetic modification in eukaryotic RNA,with massive recognition proteins and a complex regulatory network,which can play a role in gene expression regulation and cell fate determination.Increasingly studies have shown that m6A can not only regulate gene expression at the post-transcriptional level but also extensively participate in the regulation of gene transcription.m6A plays direct role in regulating transcription by affecting RNA polymerase Ⅱ promoter-proximal pausing and R-loop formation,it also plays indirect role through influencing chromatin structure and activity.This paper briefly introduces m6A and m6A co-transcriptional writing,and discusses the regulatory role of m6A in gene transcription and its relevant biological functions.
