卵巢癌是致死率较高的妇科恶性肿瘤,早期难以发现,并易复发和耐药.沉默信息调节因子1(silent information regulator 1,Sirt1)是一种烟酰胺腺嘌呤二核苷酸依赖性脱乙酰酶,在广泛的生物学事件中发挥关键作用,包括细胞凋亡、氧化应激、炎症和肿瘤的发生发展.本文综述了Sirt1在卵巢癌中表达水平明显升高,可通过自噬、氧化应激、免疫应答、血管生成及信号分子途径来促进卵巢癌的发生发展,包括卵巢癌细胞增殖、凋亡、侵袭、转移、化疗耐药及预后,以期为卵巢癌的临床治疗开辟新治疗途径提供理论基础.
The role of Sirt1 in ovarian cancer
Ovarian cancer is the gynecological maglignancies with the high mortality,which is difficult to detect in the early stage,and is easy to relapse and develop drug resistance.Silent information regulator 1(Sirt1)is a nicotinamide adenine dinucleotide-dependent deacetylase that plays critical roles in a broad range of biological events,including cell apoptosis,oxidative stress,inflammation,occurrence and development of tumor.This paper reviews that Sirt1 expression is dramatically elevated in ovarian cancer,which can facilitate the occurrence and development of ovarian cancer by regulating the autophagy,oxidative stress,immune response,angiogenesis and signaling molecules,including ovarian cancer cell proliferation,apoptosis,invasion,migration,chemoresistance and outcomes,in order to provide theoretical basis to open a new avenue for clinical treatment of ovarian cancer.
