Single-molecule sequencing is a technology in which each nucleic acid molecule can be individually sequenced without polymerase chain reaction amplification.It has the advantages of long reading length,high throughput,high sequencing speed,and is a major innovation in the history of nucleic acid sequencing technology.Single-molecule real-time sequencing and nanopore single molecule sequencing are two typical representatives of single-molecule sequencing technology.In this paper,the characteristics,principles and basic process of sequencing of these two technologies were summarized,and the progress of single-molecule sequencing technology in genome and transcriptome research of food crops,economic trees and medicinal plants were summarized,in order to provide reference for the research on molecular biology of agriculture and forestry related plants.
